诱导人骨髓间充质干细胞分化为角膜上皮细胞的实验研究
Study on differentiation of human bone marrow mesenchymal stem cells into corneal epithelial cells
目的：探讨人骨髓间充质干细胞分化为角膜上皮细胞的可行性及诱导条件；方法：常规密度梯度离心-贴壁法筛选、扩增人骨髓间充质干细胞，CellTrakerTMGreen CMFDA标记细胞；组织块法培养兔角膜缘上皮细胞。将两种细胞共培养1周，观察骨髓间充质干细胞表达角膜上皮细胞特殊分化标志keratin 3的情况；结果：经过1周共培养的诱导，骨髓间充质干细胞开始表达较低水平的keratin 3；结论：骨髓间充质干细胞具有分化为角膜上皮细胞的 潜能。
Purpose: To study the differentiatial ability of human bone marrow mesenchymal stem cells (hMSCs) to cornea epithelial cells. Method: hMSCs was isolated ex-vivo by density-gradient centrifuge followed by adhension to the flask, and expand to P3. Before induction, hMSCs was labbeled by CellTrakerTMGreen CMFDA. Rabbit corneal limbal epithelial cells was obtained by limbal tissue culture. Co-culture these two cells by cell culture insert for one week. Result: after one-week co-culture, hMSCs begin express the cornea epithelial cells-specific marker-keratin 3/12; Conclusion: hMSCs has the potentiality to differentiate into corneal epithelial cells.
