籽用南瓜多糖提取、表征及抗氧化活性研究

籽用南瓜种子被用于炼油或食品生产,果肉是其农业副产物,本文利用低共熔溶剂提取法从籽用南瓜果肉中提取得到一种南瓜多糖PPFS,经离子交换与凝胶纯化后得到籽用南瓜多糖PPFS-3。对PPFS-3进行了详细的结构表征并进行了体外抗氧化活性研究。结果表明,PPFS-3分子量为2.02x105Da,主要由岩藻糖、阿拉伯糖、鼠李糖、半乳糖、葡萄糖、木糖、甘露糖、半乳糖醛酸、葡萄糖醛酸九种单糖组成,摩尔比为1.2 : 7.2 : 9.85 : 7.16 : 1.16 : 0.74 : 0.52 : 71.21 : 1.41。首次对籽用南瓜多糖进行了一级结构表征,半乳糖、半乳糖醛酸、鼠李糖组成了PPFS-3的主链结构,阿拉伯糖主要存在于PPFS-3的支链结构上,说明PPFS-3是一种结构复杂的酸性多糖。抗氧化活性实验结果表明,PPFS-3具有较好的抗氧化活性且效果具有剂量依赖性,有作为食品,化妆品中添加剂的潜力。

In this paper, seed-used pumpkin seeds are primarily utilized for oil extraction or food production, while the flesh is an agricultural byproduct. In this study, a polysaccharide PPFS was extracted from seed-used pumpkin flesh using a deep eutectic solvent (DES) method. PPFS was further purified via ion exchange and gel filtration chromatography to obtain PPFS-3. Comprehensive structural characterization revealed that PPFS-3 has a molecular weight of 2.02×10? Da and is composed of nine monosaccharides: fucose, arabinose, rhamnose, galactose, glucose, xylose, mannose, galacturonic acid,and glucuronicacid, with a molar ratio of 1.2:7.2:9.85:7.16:1.16:0.74:0.52:71.21:1.41. This study represents the first primary structural characterization of polysaccharides from seed-used pumpkin. The backbone of PPFS-3 primarily consists of galactose, galacturonic acid, and rhamnose, while arabinose predominantly exists in its branched chains, indicating that PPFS-3 is a structurally complex acidic polysaccharide. Antioxidant assays demonstrated that PPFS-3 exhibits significant dose-dependent antioxidant activity, suggesting its potential as a functional additive in food and cosmetic industries.
