目的:探讨有氧运动和饮食干预对肥胖小鼠血清Visfatin和内脏脂肪组织Visfatin mRNA水平的影响,及其与氧化应激(OS)的关系。方法:C57BL/6J雄性小鼠50只作为研究对象,高脂饮食造成肥胖模型后,给予有氧运动和饮食控制干预,继续喂养8周后,酶联免疫法(ELISA)测定血清中的甘油三酯(TG)和胆固醇(CHO)含量,血清中总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-px)、丙二醛(MDA)及Visfatin的含量,RT-PCR方法测定内脏脂肪中Visfatin mRNA的水平。结果:(1)肥胖模型各组的血清Visfatin水平均高于普通膳食对照组;(2)饮食干预会下调内脏脂肪Visfatin mRNA水平;有氧运动干预后,血清Visfatin水平降低,内脏脂肪Visfatin mRNA水平升高;(3)有氧运动和饮食干预可以提高肥胖机体的抗氧化能力;有氧运动可以下调肥胖机体的氧化应激水平;(4)氧化应激和Visfatin水平有显著的相关性且呈正相关(r值分别为0.505、0.627、0.520,P<0.05)。结论:有氧运动和饮食干预可降低肥胖机体的Visfatin水平和氧化应激水平,并增强机体的抗氧化能力,减少肥胖所造成的氧化应激损伤。

Objective:To study the differences of oxidative stress (OS) on obesity mice separately intervened by aerobic exercise and diet control, and the relationship with visfatin levels of visceral adipose tissue (VAT) and serum. Methods:Fifty C57BL/6J male mice were used as the research object, high-fat diet was used to establish obesity model, aerobic exercise and diet control were regarded as intervention. After eight weeks, levels of triglyceride (TG), total cholesterol (CHO), T-SOD, GSH-px, MDA and visfatin levels in serum were detected by ELISA; visfatin mRNA levels in VAT were determined by RT-PCR. Results: (1) Visfatin levels in obesity model were higher than the normal diet group. (2) Diet control can down-regulate the levels of visfatin mRNA in VAT; visfatin levels in serum lower but visfatin mRNA in VAT were higher than the obesity group after exercise intervention.(3) Aerobic exercise and diet control can improve antioxidant capacity; aerobic exercise can down-regulate the levels of oxidative stress.(4) Oxidative stress was positively related to visfatin levels. Conclusion: Aerobic exercise and diet control can down-regulate the levels of OS and visfatin in obese mice and improve their antioxidant capacity.
