活化AMPK促进骨骼肌蛋白质降解过程中FoxO3 mRNA的表达
Effects of AMPK Activation on the Expression of FoxO3 mRNA in the Skeletal Muscle Protein Degradation in Rat
目的:通过给大鼠注射AMPK激动剂AICAR,提高大鼠骨骼肌中AMPK的活性,观察FoxO3 mRNA表达量的变化,探讨骨骼肌蛋白质的降解机制。方法: 24只雄性SD大鼠,8月龄,180~200g,根据用药后的取材时间分4组:AICAR注射后1 h组、2 h组、7 h组,生理盐水对照组。采用同位素技术测定腓肠肌中AMPK活性变化,采用荧光定量PCR技术, 测定腓肠肌中MuRF1、MAFbx、FoxO3基因表达量的变化。结果: AICAR注射后1 h组、2 h组、7 h组,AMPK活性与对照组相比显著性升高(P<0.05或 P<0.01),AICAR注射后7 h组,AMPK活性开始下降;AICAR注射后1 h组、2 h组,MAFbx mRNA、MuRF1mRNA表达量与对照组相比非常显著性升高(P<0.01),分别升高2.45、2.23倍和2.67、2.30倍;AICAR注射后7h组,MuRF1 mRNA表达量显著性升高(P<0.05),升高2.01倍;FoxO3mRNA表达量在AICAR注药组与对照组之间没有显著差异。结论:本研究认为大鼠一次性注射AICAR,提高AMPK活性,可能不是通过促进FoxO3 mRNA的表达,促进骨骼肌蛋白质的降解。
Purpose: By injecting AICAR, an AMPK excitant, into SD rats to boost the activity of AMPK in the skeletal muscles of the rats, the authors probed into the effects of FoxO3 mRNA changes on the degradation of proteins in skeletal muscles. Methods: Rats was divided into a bout AICAR injection, with a subcutaneous injection (AICAR; 0.5 mg/g body wt; 75 mg AICAR/ml saline), or an equivalent volume of normal saline as control. Gastrocnemius samples were collected at 1hour, 2 hours and 7 hours after AICAR injection. MAFbx mRNA, MURF-1 mRNA, FoxO3 mRNA was determined by Realtime PCR. AMPK activity was determined by [γ-32P] ATP as P donor and SAMS peptide as substrate. Results: AMPK activity were significantly increased(P<0.05)at post-inject 1 hour, 2 hours and 7 hours, then decreased at post-inject 7 hours. AICAR injection increased mRNA Expression of MuRF1(2.67-fold,2.30-fold)and MAFbx(2.45-fold, 2.23-fold) at post-inject 1 hour and 2 hours, respectively. MuRF1 mRNA were significantly elevated(P<0.05), whereas MAFbx mRNA has increased, but no significant differences at post-inject 7 hours; FoxO3 mRNA has not changed at post-inject 1 hour, 2 hours and 7 hours. Conclusion: Rat with AICAR injection may increase AMPK activity and promote the protein degradation,whereas the increase of the AMPK activity may not change FoxO3 mRNA expression. Therefore foxO3 mRNA content may be dissociated from changes in skeletal muscle protein degradation.
