不同循环载荷量对兔骨腱结合部VEGF、CRP和bFGF的影响
The Effect of Different Cycle Load on The Expression of VEGF, CRP and bFGF in Rabbit Bone-Tendon Junction
摘要
目的:研究骨腱结合部过度使用性损伤发生过程中VEGF、CRP和bFGF的变化规律,为探索骨腱结合部过度使用性损伤的发病机制提供依据。方法:45只成年雌性新西兰大白兔,按循环载荷量的不同随机分成2H(小时)、4H、6H、12H和24H组。各组完成相应的循环载荷量后,处死、取材、常规组织学处理,切片,行HE染色及VEGF、CRP 和bFGF的免疫组化染色。采用MetaMorph Premier Offline对各组间组织形态计量,SAS6.1.2进行统计分析处理,显著性水平为P<0.05。结果:在骨腱结合部,随着循环载荷量的增加,VEGF阳性表达率显著增加;CRP阳性表达率未出现显著变化;bFGF的表达呈现先增加后降低的趋势。结论 骨腱结合部的血管增生和修复因子表达降低是骨腱结合部过度使用性损伤生的机制之一;炎症与过度使用性损伤发生之间的关系有待于进一步研究。
Abstract
Objective: the purpose of this study was to evaluate the effect of different cycle loads on expression of vascular endothelial growth factor (VEGF)、C-reactive protein (CRP)、basic fibroblast growth factor(bFGF) in patellar bone-tendon junction. Methods: 45 adult female New Zealand rabbits, according to the different cycle load randomly divided into 2H (hours), 4H, 6H, 12H and 24H group. The tendons at insertion sites were harvested, routine histologic processing and sections were stained with H&E, immunostained with antibodies directed against VEGF, VEGF, and bFGF. The histomorphometry were used metamorph premier offline, the statistical data were analysed by SAS6.1.2, and the level of significance for P < 0.05. Results: With the increased amount of cycle load, the rate of sections in positive-staining of VEGF was significant increase(p<0.05). The rate of sections in positive-staining of CRP did not appear significantly change(p>0.05). The optical density of the sections with bFGF showed a trend first increasing and then decreasing. Conclusion: In the bone-tendon junction, vascular proliferation and decreased expression of repair growth factor are the pathogenesis of bone tendon junction overuse injury. the relationship between inflammation and overuse injury needs a further study.关键词
