目的:探讨再负荷安静恢复和离心运动恢复对废用性骨骼肌横截面积及肌纤维类型的影响。方法:采用尾部悬吊模型。24只成年雌性SD大鼠按体重随机分为正常对照组(CON,n=6)、尾部悬吊组TS(n=6),悬吊14天后解悬吊自然恢复组(NR,n=6)和悬吊14天后离心运动恢复组(ER,n=6)。CON和TS组在14天后取材;NR和ER在28天取材。取大鼠腓肠肌作异染性染料ATPase染色。结果:尾部悬吊14天后与对照组相比,大鼠腓肠肌I型和IIa肌纤维的组成百分比显著下降(p<0.01);IIb纤维的组成百分比显著增加。Ⅰ型肌纤维横载面积(CSA)减小最为明显,较CON组减少42.2%(P<0.01);Ⅱa型肌纤维CSA较少30.0%;Ⅱb型肌纤维减少了17.9%(p<0.01)。再负荷进行14天恢复后,离心运动能使I型、IIa型和IIb型肌纤维的组成百分比和横截面积都恢复到正常甚至超过对照水平(p>0.05);自然恢复组能使I型肌纤维的组成百分比恢复到对照组水平(p>0.05),使IIa型和IIb型肌纤维CSA恢复到正常对照水平。结论:尾部悬吊造成骨骼肌废用性肌萎缩,与自然恢复相比,离心运动能较好使废用性萎缩的骨骼肌的面积和肌纤维组成百分比得到恢复。

Objective To observe the effects of eccentric exercise in counteracting the gastroenemius atrophy and muscle fibre types transformation induced by simulated weightlessness. Method Thirty female Sprague-Dawley rats were randomly assigned to three groups: control(CON), two-week tail-suspension 1 and 2(TS1, n=6&TS2, n=12). After two weeks, the control and TS1’s gastroenemius were dissected and weighed. The TS2 were continued to randomly assign to two groups: normal rehabilitation group(NR,n=6), eccentric exercise rehabilitation group(ER,n=6). After two weeks’ rehabilitation , gastroenemius was sectioned with cryostat and stained with ATPase staining. The cross sectional area (CSA) and the proportion of typeⅠ, IIa and IIb fibers were measured using Leica image analysis system..Result In TS, wet weights of the gastroenemius, CSA of typeⅠand IIb fibers, and proportion of typeⅠand IIa fibers decreased obviously, as compared with those rats in CON (P<0.01). Whereas in type IIb, the proportion were increased significantly compared with those rats in CON(p<0.01).Under 14 days rehabilitation with different condition, eccentric exercise traning promoted the CSA and proportion of typeⅠ, IIa and IIb fibers recovering to normal levels(p>0.05),whereas normal rehabilitation only restored the proportion of type I to normal levels but not the CSA.Conclusion eccentric exercise training fully promoted the the CSA and relative proportion of typeⅠ, IIa and IIb fibers in gastroenemius recovering from atrophy induced by simulated weightlessness for 14 daysin rats.
