目的:人类线粒体基因(mtDNA)的分子遗传学特征与机体的运动能力密切相关,研究前期发现7个多态位点的分布频率与普通人之间有极显著性差异(p<0.01),但这些位点变异对运动员的有氧运动能力是否有影响,还没有证实。因此本研究拟探讨mtDNA多态性与长跑运动员有氧耐力指标的关联性。方法:79名女子长跑运动员,运动主项为一级运动员以上的5/10km或马拉松运动,北方汉族,均知情同意。收集受试者静脉血提取全血DNA,进行微测序反应对受试者mtDNA的7个多态位点(mt235、mt514-515、mt16183、mt16290、mt16319、mt8794和mt12338)进行基因分型。采用递增负荷跑台方案测试受试者有氧耐力指标,包括无氧阈心率(HRAT)、无氧阈跑速(VAT)、无氧阈摄氧量(VO2AT)、无氧阈摄氧量/体重(VO2AT/Kg)、最大摄氧量(VO2max)、最大摄氧量/体重(VO2max/Kg)等指标数据。结果:通过关联分析,没有发现受试者有氧耐力指标与mt235A/G、mt514-515CA/--、mt16183T/G、mt16290C/T、mt16319C/T、mt8794G/A和mt12338T/C显著关联,说明这7个多态位点不是长跑运动员有氧耐力指标的决定因素。结论:本研究没有找到mtDNA多态位点与长跑运动员有氧耐力指标的统计学差异,表明运动能力并非这些单个多态位点决定,可能是其它因素共同作用的结果

Objective: The aerobic capacity is closely related to the molecular genetics of human mitochondrial DNA (mtDNA), and the frequencies distribution of seven loci of mtDNA were significant difference were found in previous studies (p<0.01) between long-distance athletes and controls. Therefore, this study aims to explore the relationship between mtDNA polymorphism and aerobic endurance index of long distance runners. Methods: 79 female long-distance runners, the movement of the main item for the first class athletes over the 5/10km or marathon, belonging to the northern Han population, were informed consent. Blood samples were collected from the subjects to extract whole blood DNA, and 7 polymorphic loci (mt235, mt514-515, mt16183, mt16290, mt16319, mt8794 and mt12338) of the polymorphic loci of the subjects were performed by micro sequencing. The oxygen endurance index, including anaerobic threshold heart rate (HRAT), anaerobic threshold running speed (VAT), anaerobic threshold intake oxygen (VO2AT), anaerobic threshold photo oxygen/weight (VO2AT/Kg) of maximum oxygen uptake (VO2), maximum perturbation oxygen/body weight (VO2max/Kg) and so on, were tested by increasing load treadmill protocol testing. Results: through correlation analysis and found no oxygen endurance index and mt235A/G, mt514-515CA/--, mt16183T/G, mt16290C/T, mt16319C/T, mt8794G/A and mt12338T/C significantly related subjects, indicating that the determinants of endurance index of the seven polymorphic loci is not long distance runners. Conclusion: This study did not find mtDNA polymorphism and long distance runners statistical differences in oxygen endurance index, shows that the movement is not the single polymorphic loci decision, may be the result of other factors.
