Effects of aerobic exercise in different frequencies on the vascular reactivity of the superior mesenteric arteries

目的:研究不同频率的有氧运动对大鼠离体肠系膜上动脉反应性的影响,并探讨其可能的平滑肌钾通道机制。方法:选用8周龄健康雄性Wistar大鼠18只,随机分成三组:安静对照组(SED)、低频有氧运动组(EX-3d/w)和高频有氧运动组(EX-5d/w)。运动方案为12周跑台运动,坡度0 ,20m/min, 60min/day。制备去内皮离体肠系膜上动脉血管环,用其测定各组对去甲肾上腺素(NE)、血管紧张素Ⅱ(AngⅡ)、非特异性K+通道阻断剂四乙胺(TEA)、大电导钙激活K+(BKCa)通道阻断剂Iberiotoxin(IbTX)以及BKCa通道开放剂NS1619的反应性变化。结果:各组血管对NE(10-9~3×10-5mol/L)有浓度依赖性收缩,有氧运动组张力增加幅度降低,并且EX-5d/w张力增加幅度小于EX-3d/w;AngⅡ(3×10-7mol/L)使血管张力增加,与SED相比,有氧运动组增加幅度明显减小,但EX-3d/w和EX-5d/w之间无显著性差异;TEA(3×10-3M)和IbTX(3×10-8M)均诱发血管张力增加,增加幅度EX-5d/w > EX-3d/w > SED;NE(10-5M)引起血管收缩达到最大时,给予NS1619(10-9~10-6M)可诱发血管呈浓度依赖性舒张,有氧运动组舒张反应显著增大,并且EX-5d/w舒张幅度大于EX-3d/w。结论:有氧运动可以诱发大鼠肠系膜上动脉的反应性改变,且呈频率依赖性,其中血管平滑肌BKCa通道起着重要作用。

Objective: To study the effect of aerobic exercise in different frequencies on the vascular reactivity of the superior mesenteric arteries and to explore the possible mechanisms. Method: 18 male Wistar rats (8w) were randomly divided into three groups: sedentary control group (SED), low frequency training group (3days per week, EX-3d/w), high frequency training group (5days per week, EX-5d/w). The contractile responsiveness of the superior mesenteric arteries of all groups to norepinephrine (NE), angiotensinⅡ(AngⅡ), nonspecific K+ channel blocker tetraethylammonium (TEA), large conductance Ca2+-activated K+ channel (BKCa ) blocker Iberiotoxin (IbTX) and BKCa opener NS1619 were examined. Results: NE (10-9~3×10-5M) caused a significantly increase of vascular tone in a concentration-dependent manner in all groups, while the increase amplitude was less in EX-5d/w group than in EX-3d/w group than in the SED group. AngⅡ(3×10-7M) could produce a sustained tension increase in all three groups, which was more significant in exercise groups than in SED group. TEA (3×10-3M) and IbTX (3×10-8M) could induce arterial tension increase, too. The maximal increase of vascular tone was EX-5d/w > EX-3d/w > SED. The dilation elicited by NS1619 (10-9~10-6M) on NE-induced contraction was more significant in exercise groups (EX-3d/w and EX-5d/w) than in SED group. Conclusion: aerobic exercise training can change the vascular reactivity in superior mesenteric arteries in a frequency-dependent manner, in which BKCa channels of the vascular smooth muscle cells play an important role.
