肾细胞癌特异性核酸适体YTX-2的识别及其在早期诊断中的应用

肾细胞癌(renal cell carcinomas,RCC)是肾癌的主要类型,起源于肾小管上皮细胞,占肾脏恶性肿瘤的90%左右。肾细胞癌早期检查困难,错过最佳治疗时期且可能发生转移。利用肿瘤细胞独特的分子特征(如膜蛋白的差异表达)进行靶向治疗,为精确消灭肿瘤提供了有前景的替代方案,同时减少副作用。核酸适体(Aptamer)是能与靶标特异性结合的单链DNA或RNA,具有易合成、低成本、免疫原性低等优势,被称为"化学抗体",现已被应用于癌症的诊断与治疗。本文运用序列已知的Swan Library快速获得靶向肾细胞癌的核酸适体YTX-2,并对其进行性质表征,进一步的流式细胞术和共聚焦实验证明在37℃下YTX-2能够内化进入肾细胞癌细胞,且YTX-2能够特异性识别肾细胞癌组织。综上所述,核酸适体YTX-2有望成为肾细胞癌早期诊断和靶向治疗的新型工具。

Renal cell carcinoma (RCC), originating from renal tubular epithelial cells, represents the predominant form of kidney cancer, accounting for approximately 90% of renal malignancies. Early detection of RCC remains challenging, often leading to missed optimal treatment windows and potential metastasis. Targeted therapy utilizing unique molecular features of tumor cells, such as differential expression of membrane proteins, offers a promising alternative for precise tumor elimination while minimizing adverse effects. Aptamers, single-stranded DNA or RNA molecules capable of specific target binding, are termed "chemical antibodies" due to their advantages of easy synthesis, low cost, and minimal immunogenicity, and have been applied in cancer diagnosis and therapy. This study employed the sequence-known Swan Library to rapidly obtain the RCC-targeting aptamer YTX-2, followed by comprehensive characterization. Flow cytometry and confocal microscopy experiments demonstrated that YTX-2 could internalize into RCC cells at 37 C and specifically recognize RCC tissues. Collectively, the aptamer YTX-2 holds promise as a novel tool for early diagnosis and targeted therapy of renal cell carcinoma.
