牦牛骨源抗炎肽改善皮肤炎症的作用与机制
皮肤炎症可诱发多种皮肤疾病。抗炎肽因其疗效显著且不易引起明显副作用,已被广泛应用于皮肤问题治疗。在前期研究中,我们从牦牛骨胶原中筛选出六种抗炎肽,但其对皮肤炎症的作用尚未明确。为拓展牦牛骨胶原肽的应用领域,本研究基于脂多糖诱导的HaCaT细胞炎症模型,探究了所筛抗炎肽的作用效果及机制,并通过检测多环境因素下的肽稳定性评估其应用潜力。结果显示:当浓度为100 μM时,GPAGPSGPAGKDGR(Seq2)对IL-6、IL-1β、TNF-α和NO的抑制率分别为85.71 ± 1.40%、78.10 ± 4.02%、80.60 ± 1.34%和94.79 ± 0.48%;与模型组相比,Seq2处理组HaCaT细胞中p-P65/t-P65比值和p-IκB/t-IκB比值分别降低约32.11%和39.39%。此外,Seq2在50℃以下表现出良好的热稳定性,pH值、光照、金属离子及典型加工助剂对其稳定性均无显著影响,在特定作用范围内能保持初始浓度90%左右。研究表明:牦牛骨胶原源肽Seq2通过调控NF-κB信号通路改善HaCaT细胞炎症,且具有优异的稳定性。本研究为食源性抗炎肽的开发提供了新思路,也为牦牛骨蛋白的深入探索指明了方向。
Skin inflammation can contribute to various skin disorders. Anti-inflammatory peptides have been widely used in treating skin conditions due to their strong therapeutic effects and minimal side effects. In our previous study, six anti-inflammatory peptides were identified from yak bone collagen, though their effects on skin inflammation remained unexplored. To further investigate the potential applications of these peptides, we examined the anti-inflammatory effects and mechanisms of the selected peptide, GPAGPSGPAGKDGR (Seq2), using a lipopolysaccharide (LPS)-induced HaCaT cell inflammation model. Additionally, we assessed the peptide\'s stability under various environmental conditions to evaluate its practical applicability.At a concentration of 100 μM, Seq2 significantly inhibited the production of key inflammatory mediators, with suppression rates of 85.71 ± 1.40% for IL-6, 78.10 ± 4.02% for IL-1β, 80.60 ± 1.34% for TNF-α, and 94.79 ± 0.48% for NO. Furthermore, Seq2 treatment reduced the p-P65/t-P65 and p-IκB/t-IκB ratios in HaCaT cells by approximately 32.11% and 39.39%, respectively, compared to the model group, suggesting modulation of the NF-κB signaling pathway.Stability tests revealed that Seq2 remained highly stable at temperatures below 50 C, and its structure was largely unaffected by pH variations, light exposure, metal ions, or common processing aids, retaining around 90% of its initial concentration under these conditions.These findings demonstrate that Seq2, a yak bone collagen-derived peptide, exhibits excellent stability and effectively alleviates inflammation in HaCaT cells by regulating the NF-κB pathway. This study expands the potential applications of food-derived anti-inflammatory peptides and provides new insights for future research on yak bone proteins.
