RG介导KAR/SL调控种子萌发的分子机制探究

Karrikins(KARs)与独脚金内酯(strigolactones,SLs)同样具有丁烯酸内酯结构,并且同样具有促进种子萌发的能力。但是,KAR信号和SL信号调控种子萌发的分子机制均未得到清晰的阐明。前期发现了拟南芥基因AtDRG与水稻穗萌基因OsSDR4同源,且通过对drg smax1双突突变体的表型观察发现其恢复了smax1突变体的强萌发表型,表明DRG是Karrikin通路下游调控种子萌发的关键因子,但是其具体分子机制有待研究。因此,本文通过酵母双杂、荧光酶素互补实验、双分子荧光实验、GST-pull down等互作实验证明,DRG能够与KAI2和D14以不依赖激素小分子的方式互作。进一步分析发现,KAI2、D14与DRG互作增强了DRG的转录抑制活性,从而调控了种子萌发。该研究为了揭示DRG响应KAR信号,进而转导信号至下游靶标,最终作用于种子萌发的分子机制奠定了基础。

Karrikins (KARs) share the butenolide structure with strigolactones (SLs) and similarly possess the ability to promote seed germination. However, the molecular mechanisms by which KAR and SL signaling regulate seed germination remain unclear. Previous studies identified that the Arabidopsis gene AtDRG is homologous to the rice pre-harvest sprouting gene OsSDR4. Phenotypic observations of the drg smax1 double mutant revealed a restoration of the strong germination phenotype seen in the smax1 mutant, indicating that DRG is a key downstream factor in the Karrikin pathway regulating seed germination, though its precise molecular mechanism requires further investigation. Therefore, this study employed yeast two-hybrid, luciferase complementation assay, bimolecular fluorescence complementation, and GST-pull down interaction experiments to demonstrate that DRG can interact with KAI2 and D14 in a hormone molecule-independent manner. Further analysis revealed that the interaction of KAI2 and D14 with DRG enhances DRG\'s transcriptional repression activity, thereby regulating seed germination. This research lays the foundation for elucidating the molecular mechanism by which DRG responds to KAR signaling, transduces the signal to downstream targets, and ultimately influences seed germination.
