硫酸软骨素蛋白聚糖类似物微球的制备与表征
为模拟细胞外基质中天然硫酸软骨素蛋白聚糖的"梳型"结构,本研究首先合成了低接枝度的甲基丙烯酰化明胶,并以其为核心蛋白,理性设计了一种硫酸软骨素蛋白聚糖类似物:低分子量硫酸软骨素-甲基丙烯酰化明胶(low molecular weight chondroitin sulfate-methacryloyl gelatin ,GMCS)。本文通过傅里叶红外光谱、Zeta电位分析仪和流变仪等仪器研究了不同分子量硫酸软骨素的接枝对于甲基丙烯酰化明胶结构及物理性能的影响;通过细胞毒性试验筛选GMCS,成功用于制备支持神经再生的微球。结果表明,接枝硫酸软骨素会提高甲基丙烯酰化明胶的亲水性与电负性,当GMCS所接枝的硫酸软骨素糖链分子量在1~3 kDa时,GMCS可以保留其甲基丙烯酰化明胶主链的可光固化特性,并且对神经元生长表现出促进作用。通过微流控技术制备的GMCS水凝胶微球大小均匀,表面光滑,在组织工程中具有广阔应用前景。
In order to simulate the comb-like structure of natural chondroitin sulfate proteoglycan in extracellular matrix, this study first synthesized methacryloyl gelatin with low grafting degree, and rationally designed a chondroitin sulfate proteoglycan analog: low molecular weight chondroitin sulfate-methacryloyl gelatin (GMCS) with it as the core protein. In this paper, the effects of grafting chondroitin sulfate with different molecular weights on the structure and physical properties of methacrylated gelatin were studied by Fourier infrared spectroscopy, Zeta potential analyzer and rheometer. GMCS were screened by cytotoxicity test and successfully used to prepare microspheres supporting nerve regeneration. The results show that the grafting of chondroitin sulfate can improve the hydrophilicity and electronegativity of methacrylated gelatin. When the molecular weight of chondroitin sulfate grafted by GMCS is 1 ~ 3 kDa, GMCS can retain the photocurable characteristics of the main chain of methacrylated gelatin and promote the growth of neurons. GMCS hydrogel microspheres prepared by microfluidic technology have uniform size and smooth surface, and have broad application prospects in tissue engineering.
