光甘草酮对皮肤黑色素合成的抑制作用及其机理研究
目的:探究光甘草酮(Glabrone)抑制细胞黑色素合成的作用及其机理。方法:以B16细胞为模型,分析光甘草酮对黑色素合成的抑制效果;通过细胞和体外酪氨酸酶活性实验,探究其抑制途径;为探索其作用机制,以DPPH自由基清除实验和UVB诱导的细胞活性氧清除实验评估其抗氧化能力,检测B16细胞内酪氨酸酶、小眼畸形相关转录因子、环磷腺苷效应元件结合蛋白的mRNA分析其生理效应,最后,通过UVB诱导的小鼠皮肤色素沉着模型验证光甘草酮干预黑色素合成的效果。结果:光甘草酮可显著降低B16细胞内黑色素含量,抑制细胞和体外酪氨酸酶活性,抑制率分别为29.7%和30%;有效清除DPPH自由基,降低细胞内活性氧水平,下调MITF、TYR、CREB基因表达,总体效果优于光甘草定。动物实验表明,光甘草酮可显著减少UVB诱导的皮肤色素沉着。结论:光甘草酮通过抑制酪氨酸酶表达和活性,在细胞和动物水平上实现与光甘草定相当的黑色素调控效果。其机制可能与调节氧化应激通路、影响PKC-PKA-CREB信号传导、下调MITF和TYR基因表达有关。
Objective: To elucidate the inhibitory effect of Glabrone on melanogenesis and its underlying mechanisms. Methods: The inhibitory effects of Glabrone on melanin synthesis was evaluated by the B16 cell model. Subsequently, to understand the mechanism, the tyrosinase activity on B16 cell and enzyme assay were used. Next, the antioxidant capacity of Glabrone was assessed by the DPPH scavenging assay and the UVB induced intracellular oxygen species (ROS) scavenging assay. To explore the physiological process of Glabrone, qRT-PCR was used to evaluate the mRNA expression of tyrosinase (TYR), microphthalmia-associated transcription factor (MITF), and cAMP response element-binding protein (CREB) on B16 cells. Finally, the melanogenesis inhibitory effect of Glabrone on C57BL/6J mice induced by UVB irradiation was declared. Results: Glabrone significantly reduced the melanin synthesis in B16 cells, inhibited both intracellular and extracellular tyrosinase activities, with inhibition rates of 29.7%, and 30%, respectively. Compared with Glabridin, Glabrone show a better effect on DPPH free radicals scavenging, intracellular ROS cleaning, and downregulating MITF, TYR, and CREB gene expression. The animal experiments confirmed that Glabrone effectively reduce UVB-induced skin pigmentation. Conclusion: Glabrone inhibits melanogenesis by suppressing the expression and activity of tyrosinase primarily. The mechanism of Glabrone may involves regulating oxidative stress-related pathways, modulating the PKC-PKA-CREB signaling cascade and downregulating of MITF and TYR gene expression..
