RNA结合蛋白AKAP95基因组结合位点的鉴定
蛋白激酶A的锚定蛋白AKAP95(A-kinase anchoring protein 95,AKAP95)定位于细胞核,是一种RNA结合蛋白。以往研究发现AKAP95可作为转录辅因子参与转录调控,然而对于其中的细节还知之甚少。本研究通过CRISPR-Cas9技术成功构建了内源性稳定插入的Hela(Flag-mEGFP-AKAP95)细胞系,并对其进行Flag ChIP-seq实验。结果表明AKAP95广泛地结合在基因组DNA上,尤其在基因的启动子处有明显的富集结合。motif分析得出AKAP95的结合区域富集4种转录因子(NFY、Sp1、YY1、Elk4)的保守结合motif,表明AKAP95可直接与基因组DNA结合参与转录激活调控。本研究创建的内源性AKAP95 ChIP-seq研究系统,为进一步探索AKAP95参与转录调控的生物物理机制奠定了基础。
-kinase anchoring protein 95 (AKAP95), localized in the nucleus, is an RNA-binding protein. Previous studies have found that AKAP95 participates in transcriptional regulation as a transcriptional cofactor. However, little is known about the details. In this study, we successfully constructed an endogenous expression of Flag-mEGFP-AKAP95 Hela cell line by CRISPR-Cas9 technology. We performed Flag ChIP-seq experiments on it to explore AKAP95 interaction with chromatin, and found that AKAP95 widely bound to genomic DNA, especially at the gene promoters. Motif analysis revealed that the binding promoters of AKAP95 were enriched with four transcription factors (NFY, Sp1, YY1, Elk4) binding motifs, suggesting AKAP95 may be involved in transcription activation through binding to genomic DNA. The present study lays the foundation for further exploration of the biophysical mechanism of AKAP95 involved in transcription activation.
