基于花生NAM群体产量性状的QTL定位
栽培种花生(Arachis hypogaea L.)营养价值丰富、种植面积广泛,是重要的油料作物之一。由于耕地资源有限,提高花生单产一直是花生育种工作者的首要目标。花生百果重、百仁重和出仁率是花生单株产量的直接决定因素。本研究利用课题构建的NAM群体,采用全基因组关联分析(GWAS)和连锁分析相结合的方法对调控花生百果重、百仁重和出仁率相关性状的QTL位点进行共定位研究,结果表明:3个性状在NAM群体中的相关性与在四个RIL群体中的相关性表现出一致的结果,3个性状两两之间表现出极显著正相关。广义遗传率范围在0.79-0.91之间,环境、基因型及环境与基因型互作对百果重、百仁重和出仁率3个性状的表型影响均为极显著水平。连锁分析多环境下检测到与百果重、百仁重和出仁率相关的QTL区间分别有11、41、和46个,处于Chr3、Chr7和Chr15号染色体上。关联分析3个性状的SNP位点分别有372、226、36个,都在Chr7号染色体上。连锁分析结合关联分析共定位,3个性状重叠区间分别有16、28、8个。这些重叠区间可为后续精细定位及分子标记辅助育种提供理论依据。?????
ultivated peanut (Arachis hypogaea L.) is rich in nutritional value and widely cultivated, being one of the important oil crops. Due to the limited arable land resources, increasing the yield per unit area of peanut has always been the primary goal of peanut breeders. The 100-pod weight, 100-seed weight and shelling percentage are the direct determinants of the yield per plant of peanut. In this study, the NAM population constructed by our research group was used, and the QTL loci controlling the traits of the 100-pod weight, 100-seed weight and shelling percentage were co-located by combining genome-wide association analysis (GWAS) and linkage analysis. The results showed that the correlations of the three traits in the NAM population were consistent with those in the four RIL populations, and the two traits were highly significantly positively correlated. The broad-sense heritability ranged from 0.79 to 0.91. The effects of environment, genotype and the interaction between environment and genotype on the phenotypes of the three traits were all highly significant. Linkage analysis detected 11, 41 and 46 QTL intervals related to the 100-pod weight, 100-seed weight and shelling percentage in multiple environments, respectively, which were located on chromosomes 3, 7 and 15. GWAS identified 372, 226 and 36 SNP loci related to the three traits, respectively, all on chromosome 7. The co-location of linkage analysis and GWAS identified 16, 28 and 8 overlapping intervals for the three traits, respectively.?These overlapping intervals can provide a theoretical basis for subsequent fine localization and molecular marker-assisted breeding.????
