血管内皮细胞DKK2在动脉粥样硬化中的作用机制研究
目的:旨在探究血管内皮细胞DKK2参与动脉粥样硬化形成的分子作用机制。方法:通过构建内皮细胞Dkk2特异性敲除小鼠,探究内皮细胞DKK2对于斑块形成的影响;同时在体外构建DKK2过表达的内皮细胞系,并对其进行普通转录组测序,揭示DKK2对于内皮细胞功能的调节及其机制。结果:(1)DKK2在动脉粥样硬化患者和斑块组织中高表达;(2)血管内皮细胞DKK2促进动脉粥样硬化斑块的形成并增加斑块的不稳定性;(3)DKK2显著增强内皮细胞的黏附功能。
Objective:This study aimed to investigate the molecular mechanism by which vascular endothelial cell-derived DKK2 contributes to the development of atherosclerosis.Methods:An endothelial cell-specific DKK2 knockout mouse model was established to explore the role of endothelial DKK2 in plaque formation. In parallel, a DKK2-overexpressing endothelial cell line was constructed in vitro, followed by bulk RNA sequencing to reveal the regulatory effects and underlying mechanisms of DKK2 on endothelial cell function.Results:(1) DKK2 was highly expressed in atherosclerotic patients and plaque tissues;(2) Endothelial DKK2 promoted the formation of atherosclerotic plaques and increased plaque instability;(3) DKK2 enhanced endothelial adhesion.
