两个玉米连锁群体的生育酚含量QTL定位
为解析玉米籽粒中生育酚的遗传变异机制,选取CIMBL83/GEMS41与TY1/GEMS41两个玉米连锁群体作为遗传材料。基于全基因组标记构建遗传图谱,结合高效液相色谱测定玉米籽粒生育酚含量作为表型,进行QTL定位。主要结果如下:在CIMBL83/GEMS41和TY1/GEMS41两个群体中分别筛选到1488和1167个多态性SNP标记并构建了遗传图谱,标记间的平均图距依次为1.26cM、1.84cM。对两个群体生育酚表型进行基本统计分析发现两个群体中生育酚各性状表型变异丰富,总生育酚含量分别在7.94~45.11 μg/g和6.25~55.21 μg/g之间,并且各性状间均具有一定的相关性。采用复合区间作图法分别对2个群体进行了QTL分析,在CIMBL83/GEMS41群体中定位到了7个QTL,TY1/GEMS41群体中定位到了12个QTL,其中共包含5个主效QTL。这些主效QTL在育种中应用可以有效提高玉米籽粒中的生育酚含量,为后续培育高生育酚含量的玉米种质提供参考。
o elucidate the genetic variation mechanisms of tocopherol in maize kernels, two maize linkage populations, CIMBL83/GEMS41 and TY1/GEMS41, were selected as genetic materials. Genetic linkage maps were constructed based on genome-wide markers, and tocopherol content in maize kernels was measured using high-performance liquid chromatography (HPLC) as phenotypic data for QTL mapping. The main findings are as follows: In the CIMBL83/GEMS41 and TY1/GEMS41 populations, 1,488 and 1,167 polymorphic SNP markers were screened respectively to construct genetic linkage maps, with average genetic distances between markers of 1.26 cM and 1.84 cM. Basic statistical analysis of tocopherol-related phenotypic traits in both populations revealed abundant phenotypic variation, with total tocopherol content ranging from 7.94-45.11 μg/g in CIMBL83/GEMS41 and 6.25-55.21 μg/g in TY1/GEMS41, along with correlations among traits. QTL analysis using composite interval mapping (CIM) identified 7 QTLs in the CIMBL83/GEMS41 population and 12 QTLs in the TY1/GEMS41 population, including 5 major QTLs (PVE > 10%). The application of these major QTLs in breeding programs can effectively enhance tocopherol content in maize kernels, providing a valuable reference for developing high-tocopherol maize germplasm.
