黑莓的降脂活性及其入血成分分析

目的探讨黑莓原液与黑莓冻干粉对肥胖小鼠的降脂作用及其血清代谢物的变化,分析其降脂机制。方法采用高脂饮食诱导肥胖小鼠模型,分别给予黑莓原液和黑莓冻干粉干预,持续6 w。通过监测小鼠的体重变化、血糖水平、葡萄糖耐量试验及肝脏病理学分析,评估其降脂效果。此外,利用超高效液相色谱-四极杆-飞行时间质谱联用(UPLC-QTOF-MSE)技术分析血清代谢产物,探讨黑莓的活性成分及其降脂机制。结果黑莓干预显著减缓了高脂饮食引起的小鼠体重增加,并改善了糖代谢紊乱。高剂量黑莓原液和黑莓冻干粉组在体重、血糖和葡萄糖耐量方面均表现出显著改善。成分分析显示,黑莓冻干粉富含多种活性成分,尤其是糖苷类、氨基酸类和黄酮类,而黑莓原液主要含有糖类和氨基酸。血清代谢产物分析显示,黑莓冻干粉通过糖类、氨基酸及萜类等多种成分调节脂质代谢,表现出较强的降脂作用,而黑莓原液则可能主要通过改善糖代谢及抗氧化作用发挥其降脂效果。结论黑莓具有显著的降脂和改善糖代谢的作用,其复杂的活性成分在肥胖和代谢综合征的防治中展现出潜力,为其在相关疾病治疗中的应用提供了理论依据。

Objective To investigate the lipid-lowering effects of blackberry juice and freeze-dried blackberry powder in obese mice and analyze the changes in their serum metabolites to explore the underlying mechanisms. Methods High-fat diet-induced obese mice were treated with blackberry juice and freeze-dried blackberry powder for 6 weeks. The effects of treatment on body weight, blood glucose, glucose tolerance, and liver pathology were assessed. Additionally, serum metabolites were analyzed using UPLC-Q TOF-MSE to identify active components and their potential lipid-lowering mechanisms. Results Blackberry intervention significantly reduced weight gain induced by a high-fat diet and improved glucose metabolism. The high-dose groups of both blackberry juice and freeze-dried blackberry powder showed significant improvements in body weight, blood glucose, and glucose tolerance. Component analysis revealed that freeze-dried blackberry powder contained various active compounds, including glycosides, amino acids, and flavonoids, whereas blackberry juice primarily contained sugars and amino acids. Serum metabolite analysis indicated that freeze-dried blackberry powder regulated lipid metabolism through multiple components, such as sugars, amino acids, and terpenes, exhibiting a strong lipid-lowering effect. Blackberry juice primarily exerted its lipid-lowering effect by improving glucose metabolism and antioxidant activity. Conclusion Blackberry has significant lipid-lowering and glucose metabolism-improving effects. Its complex active components show potential for the prevention and treatment of obesity and metabolic syndrome, providing a theoretical basis for its clinical application in related diseases.
