基于上皮间质转化探究乳铁蛋白对放射性肠损伤的防护作用
目的:通过肠上皮间质转化角度探究乳铁蛋白干预对放射性肠损伤的保护作用。方法:雄性C57BL/6小鼠32只,随机分为四组,对照组(Control)、单纯照射组(10Gy)、低剂量乳铁蛋白+10Gy照射联合组(0.5 g/kg +10Gy,LLf)和高剂量乳铁蛋白+10Gy照射联合组(1.0 g/kg +10Gy,HLf)。采用X射线单次照射小鼠腹部,总剂量为10 Gy,制备放射性肠损伤小鼠模型。乳铁蛋白干预组在照射前7天至照射前1小时给予灌胃,1次/天。照射后7天处死,收集小鼠肠组织样本,进行肠功能评价及相关分子机制研究。结果:乳铁蛋白干预有效改善了小鼠的一般生命体征;在基因水平上调紧密连接结构蛋白1(ZO-1)、跨膜闭合蛋白(Occludin)和紧密连接蛋白1(Claudin-1);维持了肠上皮微绒毛长度和线粒体完整性;肠上皮间质转化的结果显示,乳铁蛋白的干预有效抑制了辐射所致的E-钙粘蛋白(E-cadherin)的下调和N-钙粘蛋白(N-cadherin)和平滑肌肌动蛋白α(α-SMA)等的上调。结论:乳铁蛋白干预可通过调节辐射所致的肠上皮间质转化,改善受照小鼠放射性肠损伤。
Objective: This study aimed to investigate the protective effect of lactoferrin intervention on radiation-induced intestinal injury by focusing on intestinal epithelial-mesenchymal transition (EMT). Thirty-two male C57BL/6 mice were randomly divided into four groups: control (Control), irradiation-only (10Gy), low-dose lactoferrin + 10Gy irradiation (0.5 g/kg +10Gy, LLf), and high-dose lactoferrin + 10Gy irradiation (1.0 g/kg +10Gy, HLf). A single abdominal X-ray irradiation was administered to establish a radiation-induced intestinal injury model. Lactoferrin intervention groups received daily oral gavage for 7 days before irradiation until 1 hour prior to exposure. Mice were euthanized 7 days post-irradiation for intestinal tissue collection, functional evaluation, and molecular analysis. Results: Lactoferrin intervention significantly improved general health status, upregulated mRNA levels of tight junction proteins (ZO-1, Occludin, Claudin-1), preserved intestinal epithelial microvillus length and mitochondrial integrity, and suppressed radiation-induced EMT by inhibiting E-cadherin downregulation and N-cadherin&α-SMA upregulation. Conclusion: Lactoferrin alleviates radiation-induced intestinal injury by modulating EMT progression.
