匍匐翦股颖悬浮细胞培养体系建立初探
匍匐翦股颖是禾本科翦股颖属多年生草本植物,属于冷季型草坪草,具有适应性强、观赏性好的优良特性,通过诱导匍匐翦股颖愈伤组织建立遗传转化体系是获得遗传性状较好的匍匐翦股颖的重要手段。植物细胞悬浮培养技术可以通过液体培养基培养植物细胞以及较小的细胞团,悬浮培养的植物细胞不受外部因素限制具有相对较短的倍增时间,且能够产生较多次生代谢物。为了建立良好稳定的匍匐翦股颖悬浮培养体系,本实验以匍匐翦股颖胚性愈伤组织为材料,初步探索了匍匐翦股颖悬浮细胞的生长增殖过程,适宜的培养环境以及对潮霉素的敏感程度,为建立生活力强,保持细胞旺盛增殖的悬浮体系和遗传转化体系提供理论支撑。结果表明,在诱导悬浮细胞前,对愈伤组织进行继代,继代2次后,可获得适宜诱导悬浮细胞的鲜亮淡黄色、质地疏松、易分散、不容易分泌黏液的颗粒状胚性愈伤组织;悬浮培养初期每3d继代换液一次,新鲜培养液与原培养液控制在2:1左右;1个月后,悬浮培养体系建立后,处于稳定状态的匍匐翦股颖悬浮细胞最佳的换液继代培养时间是8-10d一次,新鲜培养液与原培养液比例继续控制在2:1左右的条件下培养,适宜匍匐翦股颖悬浮细胞的再生;匍匐翦股颖悬浮细胞生长最佳的pH值区间在5.0-5.5之间;在进行农杆菌介导的遗传转化时,适合匍匐翦股颖悬浮细胞筛选的潮霉素浓度为15 mg/L。本研究探明了匍匐翦股颖悬浮细胞适宜的诱导条件,继代周期以及培养基适宜的pH值,完善了匍匐翦股颖悬浮细胞团再生体系。为了建立匍匐翦股颖悬浮细胞遗传转化体系,本实验确定了适合匍匐翦股颖悬浮细胞筛选的潮霉素浓度。?????

In order to establish a good and stable creeping bentgrass suspension culture system. The preliminary process about creeping bentgrass suspension cell\'s growth and proliferation was explored. This study provides theoretical support for the establishment of suspension system and genetic transformation system with strong viability and vigorous cell proliferation. The results showed that after subculture of callus for two times before the induction of suspension cells, the granular embryogenic callus with bright yellow color, loose texture, easy dispersion and hard to secret mucus could be obtained. At the initial stage of suspension culture, the subculture medium was changed every 3 days, and the fresh culture medium and the original culture medium were controlled at about 2:1, which helped to maintain the vigorous proliferation of cells. One month later, after the establishment of suspension system, the optimal subculture time of suspension cells in stable state was 8-10d. The ratio of fresh culture medium to original culture medium continued to be controlled at about 2:1, which was suitable for the regeneration of suspension cells of creeping bentgrass. The optimum pH value for the growth of suspension cells of the creeping bentgrass was between 5.0 and 5.5. 15 mg/L was suitable for Agrobacterium-mediated genetic transformation in the suspension cells of the creeping bentgrass.
