氢甲酰化反应是工业催化中最重要的反应之一。近年来,氢甲酰化反应在药物分子的构建中逐渐展现出巨大潜力。本文系统综述了氢甲酰化反应在药物合成中的应用进展,涵盖其在维生素A醋酸酯、非尼拉敏、萘普生、奥马曲拉、布洛芬、芬地林及天然产物等代表性药物中的应用实例,指出当前研究中存在的问题与挑战,并提出发展高效新型膦配体的研究方向。本文旨在为氢甲酰化反应在药物合成中的进一步研究和应用提供理论参考与实践指导。

Hydroformylation is one of the most important reactions in industrial catalytic chemistry. In recent years, hydroformylation has demonstrated considerable potential in the construction of pharmaceutical molecules. This review provides a comprehensive summary of recent advances in the application of hydroformylation in drug synthesis, highlighting representative examples including vitamin A acetate, pheniramines, naproxen, omapatrilat, ibuprofen, fendiline, and various natural products. The article also identifies current challenges and limitations in this field, and proposes the development of novel, highly efficient phosphine ligands as a promising direction for future research. This work aims to offer both theoretical insights and practical guidance for the continued application of hydroformylation in pharmaceutical synthesis.
