【目的棉花泛素结合酶基因GhRCE1调控黄萎病抗性的功能研究】棉花泛素结合酶基因GhRCE1调控黄萎病抗性的功能研究棉花泛素结合酶基因GbDIR78调控黄萎病抗性的功能研究】通过对陆地棉GhRCE1基因进行抗病表型鉴定及抗病机制解析,验证GhRCE1是否参与调控黄萎病抗性,并深入解析其作用机理。【方法】本研究利用遗传学、分子生物学等手段,对GhRCE1是否调控黄萎病抗性进行功能鉴定,并通过对活性氧相关指标及木质素含量进行测定,初步揭示了GhRCE1调控棉花黄萎病抗性的作用途径和抗病机理。【结果】GhRCE1受黄萎病菌诱导表达,抗病品种农大601和感病品种中棉所8号中GhRCE1均受黄萎病菌诱导上调表达,但农大601中的GhRCE1响应更快。GhRCE1正调控植物黄萎病抗性,沉默导致抗性显著下降,在拟南芥中过表达GhRCE1可显著提高黄萎病抗性。GhRCE1可通过影响活性氧平衡及木质素含量正向调控棉花黄萎病抗性。【结论】证实GhRCE1正调控植物黄萎病抗性,为棉花抗病机制解析提供了新视角。

Objective] Through the identification of the disease resistance phenotype and the analysis of the molecular mechanism of the GhRCE1 gene in upland cotton, it was verified whether GhRCE1 is involved in regulating the resistance to Verticillium Wilt disease, and its mechanism of action was deeply analyzed. [Method] In this study, genetic, molecular biological and other means were employed to conduct functional identification on whether GhRCE1 regulates Verticillium Wilt resistance. By measuring the relevant indicators of reactive oxygen species and lignin content, the mechanism and molecular mechanism of GhRCE1 regulating Verticillium Wilt resistance in cotton were preliminarily revealed. [Result] GhRCE1 was induced to be expressed by the Verticillium Wilt fungus. In both the disease-resistant variety ND601 and the susceptible variety CCRI8, GhRCE1 was upregulated by the Verticillium Wilt fungus, but the response of GhRCE1 in ND601 was faster. GhRCE1 positively regulates the resistance to yellow-wilt in plants. Silencing leads to a significant decrease in resistance. Overexpression of GhRCE1 in Arabidopsis thaliana can significantly improve the resistance to yellow-wilt. GhRCE1 can positively regulate the resistance of cotton Verticillium Wilt disease by influencing the balance of reactive oxygen species and the content of lignin. [Conclusion] Molecular experimental methods confirmed that GhRCE1 positively regulates the resistance of plant Verticillium Wilt disease, providing a new perspective for the analysis of the disease resistance mechanism of cotton.
