质粒拷贝数工程优化酿酒酵母中邻氨基苯甲酸甲酯的产量

邻氨基苯甲酸甲酯(Methyl anthranilate,MANT)是一种带有橙花香气的芳香族化合物,被广泛应用于工业和农业领域。本研究基于质粒拷贝数工程(Plasmid Copy Numbers,PCNs)策略,克服了酿酒酵母BJ5464-NpgA遗传操作效率低下的技术瓶颈,并利用该菌显著提高了邻氨基苯甲酸甲酯的产量。以红色荧光强度表征质粒拷贝数,通过截短博来霉素(ZeocinR)抗生素基因的启动子,同提高抗生素浓度的方式相结合,调整筛选压力,进而提高细胞中质粒的拷贝数。最终参数确定为在300 mg/L博来霉素浓度下将启动子截短至30 bp,将该参数应用于BJ5464-NpgA异源合成MANT中,HPLC分析表明,实验菌株和对照菌株的产量分别为43.57 mg/L和15.51 mg/L,产量提高1.81倍。该策略基于游离的质粒表达系统为优化酿酒酵母的异源代谢途径提供了新思路,同时可为构建微生物天然产物生产平台提供参考。

Methyl anthranilate (MANT) is an aromatic compound with an orange blossom fragrance, widely used in industrial and agricultural applications. In this study, we developed a plasmid copy number engineering (PCNs) strategy to overcome the technical bottleneck of low genetic manipulation efficiency in Saccharomyces cerevisiae BJ5464-NpgA, and achieved significant enhancement of methyl anthranilate production using this engineered strain.Using red fluorescence intensity as an indicator of plasmid copy number, we precisely modulated the selection pressure by truncating the promoter of the Zeocin resistance gene and simultaneously elevating antibiotic concentration, thereby boosting plasmid copy number. The optimal parameters were determined to be a 30 bp truncated promoter under a Zeocin concentration of 300 mg/L.When applied to the heterologous synthesis of MANT in BJ5464-NpgA, HPLC analysis revealed that the engineered strain achieved a titer of 43.57 mg/L, compared to 15.51 mg/L in the control strain, representing a 1.81-fold increase in production. This strategy,based on an episomal plasmid expression system, provides a novel approach for optimizing heterologous metabolic pathways in Saccharomyces cerevisiae, and offers valuable insights for the development of microbial platforms for natural product biosynthesis.
