银介导的自由基串联环化合成吲哚异喹啉衍生物

含氮杂环化合物是天然产物和合成化学中的一类基本化合物,在生物分子和药物结构中起着关键作用。其中稠环吲哚衍生物是极为重要的一种,它在有机合成和药物中非常重要,广泛存在于药物合成、天然产物和各种功能材料中。由于吲哚衍生物有着普遍应用,所以一直以来都受到医药和合成化学家的关注。时至今日,合成吲哚衍生化合物的方法有很多报道,也取得很大的成就,但仍然面临若干挑战。因此继续开发和发展更简单高效的合成方法有重要意义。本文主要研究一种银介导的自由基加成串联环化反应,构建稠环吲哚衍生物,进一步发展自由基加成串联环化反应在杂环合成方面的研究与应用。实验以2-芳基-N-甲基丙烯酰吲哚和α-羰基溴化物为底物,利用Ag2CO3的单电子还原特性,通过自由基加成串联环化,合成吲哚并[2, 1-a]异喹啉衍生物。体系中除了反应底物外,只需加入了银盐和溶剂,反应条件简单,且底物范围较广。

Nitrogen-containing heterocyclic compounds are a basic type of compounds in natural products and synthetic chemistry, playing a key role in the structure of biomolecules and drugs. Among them, polycyclic indole derivatives are an extremely important type. They are of great significance in organic synthesis and pharmaceuticals, and are widely present in drug synthesis, natural products, and various functional materials. Because indole derivatives have wide applications, they have always attracted the attention of medical and synthetic chemists. To this day, there have been many reports on the methods for synthesizing indole-derived compounds and significant achievements have been made, but several challenges still remain. Therefore, it is of great significance to continue to develop and advance simpler and more efficient synthetic methods. This paper mainly studies a silver-mediated radical addition tandem cyclization reaction, constructs polycyclic indole derivatives, and further develops the research and application of radical addition tandem cyclization reactions in heterocyclic synthesis. In the experiment, 2-aryl-N-methylacrylyl indole and α -carbonyl bromide were used as substrates. By taking advantage of the single-electron reduction property of Ag2CO3 and through free radical addition tandem cyclization, indole and [2, 1-a] isoquinoline derivatives were synthesized. In the system, apart from the reaction substrates, only silver salts and solvents are added. The reaction conditions are simple and the range of substrates is relatively wide.
