摘要

目的:针对CD30阳性肿瘤,开发靶向CD30的嵌合抗原受体T细胞(Chimeric antigen receptor modified T cells, CAR-T),体外探究三种CAR-T细胞对于CD30阳性肿瘤的抗肿瘤疗效。方法:通过慢病毒转染的方式构建三种CD30-CAR-T,通过流式细胞术对慢病毒转导效率、体外杀伤能力、分化表型、表面活化及耗竭表达情况等进行检测。结果:成功制备了滴度较高的慢病毒,随后成功通过慢病毒转导的方式构建形成CD30-CAR-T细胞,最终验证了其在体外展示出较好的控制CD30阳性肿瘤生长的能力。4.结论:在本研究中成功构建了CD30-CAR-T细胞,对其在体外的抗肿瘤作用进行验证,具有较好的控制CD30阳性肿瘤进展的能力,为其临床治疗提供了一个新的方向。

Abstract

Objective:To develop CD30-targeting chimeric antigen receptor-modified T cells (CAR-T cells) for CD30-positive tumors and evaluate the antitumor efficacy of three CAR-T cell types against CD30-positive tumors in vitro.Methods:Three CD30-CAR-T cell types were constructed via lentiviral transduction. Flow cytometry was used to assess lentiviral transduction efficiency,in vitro cytotoxic activity, differentiation phenotypes, and surface activation/exhaustion markers.Results:High-titer lentiviruses were successfully prepared, and CD30-CAR-T cells were generated via lentiviral transduction. These cells demonstrated robust ability to control the growth of CD30-positive tumors in vitro.Conclusion: This study successfully constructed CD30-CAR-T cells and validated theirin vitroantitumor effects against CD30-positive tumors, demonstrating promising potential for clinical translation.
