内侧前额叶皮层介导6-OHDA小鼠痛敏反应的机制研究
目的:本研究探讨内侧前额叶皮层(medial prefrontal cortex,mPFC)对6-OHDA诱导的PD(parkinson\'s disease)小鼠痛觉过敏的调控作用及机制。方法:实验选用C57BL/6J雄性小鼠,通过脑立体定位注射6-OHDA至右脑MFB(medial forebrain bundle,MFB)构建PD模型。采用von Frey和甩尾实验检测痛阈变化,悬尾、旷场及高架十字迷宫实验评估负性情绪变化。通过c-Fos免疫荧光染色方法检测mPFC脑区的c-Fos阳性神经元数目,并鉴别其神经元类型。采用化学遗传结合行为学方式检测mPFC脑区内谷氨酸能神经元疼痛相关行为的调控作用。结果:相较于Sham组,6-OHDA组小鼠痛阈显著下降,悬尾静止时间增多,旷场中心区域活动时间及高架十字迷宫开放臂活动时间减少。6-OHDA小鼠mPFC脑区的c-Fos阳性神经元数量增加,且76%与谷氨酸能神经元共定位。激活mPFC脑区谷氨酸能神经元后,6-OHDA小鼠痛阈下降,抑制则与之相反。结论:mPFC调控6-OHDA诱导的PD小鼠痛觉过敏,可能与谷氨酸能神经元异常激活相关。
Objective: This study aimed to investigate the regulatory role and mechanism of the medial prefrontal cortex (mPFC) in pain hypersensitivity in 6-OHDA-induced Parkinson\'s disease (PD) mice. Methods: Male C57BL/6J mice were used in the experiment. PD models were established by stereotactic injection of 6-OHDA into the right medial forebrain bundle (MFB). The von Frey and tail flick tests were used to detect changes in pain threshold, and the tail suspension, open field, and elevated plus maze tests were used to evaluate negative emotional changes. The number of c-Fos positive neurons in the mPFC was detected by c-Fos immunofluorescence staining, and their neuronal types were identified. The role of glutamatergic neurons in the mPFC in pain-related behaviors was detected by chemogenetic combined with behavioral methods. Results: Compared with the Sham group, the pain threshold of 6-OHDA group mice decreased significantly, the tail suspension immobility time increased, and the activity time in the center area of the open field and the open arm of the elevated plus maze decreased. The number of c-Fos positive neurons in the mPFC of 6-OHDA mice increased, and 70% of them were co-localized with glutamatergic neurons. After activating glutamatergic neurons in the mPFC, the pain threshold of 6-OHDA mice decreased. Inhibition had the opposite effect. Conclusion: The mPFC regulates pain hypersensitivity in 6-OHDA-induced PD mice, which may be related to the abnormal activation of glutamatergic neurons.
