摘要
研究真菌Aspergilllus sp. WHUF05008中的化学学成分。采用硅胶、Sephadex LH-20凝胶及半制备高效液相色谱等方法进行分离纯化,通过核磁共振、高分辨质谱及圆二色谱等方法鉴定化合物的结构。从发酵液中次生代谢产物中分离、鉴定了4个化合物,分别为coumarincarbamide (1)、isokotanins C (2)、orlandin (3)、Demethylkotanin (4)。其中化合物1为新化合物。采用MTT法测定化合物1?4对对乙酰氨基酚诱导的AML-12细胞急性肝损伤的体外肝损伤保护活性。结果显示所有化合物均能提升肝损伤细胞存活率,其中化合物1和4在5?20 μM剂量范围内均具肝保护活性,而化合物2和3仅在5 μM时有效,更高剂量(10?20 μM)则表现出毒性。
Abstract
The study investigated the chemical constituents of the fungus Aspergillus sp. WHUF05008. Separation and purification were performed using silica gel, Sephadex LH-20 gel column chromatography, and semi preparative high performance liquid chromatography (HPLC). The structures of the isolated compounds were elucidated by nuclear magnetic resonance (NMR), high resolution mass spectrometry (HR MS), and circular dichroism (CD) spectroscopy. Four secondary metabolites were isolated and identified from the fermentation broth, namely coumarincarbamide (1), isokotanin C (2), orlandin (3), and demethylkotanin (4), among which compound 1 is a new compound. The hepatoprotective activities of compounds 1?4 against acetaminophen induced acute liver injury in AML 12 cells were evaluated in vitro using the MTT assay. The results showed that all compounds increased the viability of injured cells. Compounds 1 and 4 exhibited hepatoprotective effects across the tested concentration range (5?20 μM), whereas compounds 2 and 3 showed activity only at 5?μM and displayed toxicity at higher concentrations (10?20 μM).关键词
