香菇多糖对棕榈酸诱导巨噬细胞脂质蓄积和炎症因子分泌的拮抗效应研究

摘要
[目的]:探究香菇多糖(Lentinan, LNT)对棕榈酸(Palmitic Acid, PA)诱导巨噬细胞脂质蓄积与炎症因子分泌的拮抗效应,为LNT在代谢炎症相关疾病中的应用提供实验依据。[方法]:将RAW264.7巨噬细胞分为空白对照组、PA模型组、LNT低/中/高剂量干预组。采用不同浓度PA处理RAW264.7细胞构建脂质蓄积及炎症模型,筛选最优造模浓度;后续实验中,LNT干预组提前加入对应浓度LNT预处理,再加入最优浓度PA共同培养。通过酶联免疫吸附法(ELISA)检测细胞上清液中白细胞介素-1β(IL-1β)、白细胞介素-6 (IL-6)的分泌水平,生化试剂盒检测细胞内甘油三酯(TG)、总胆固醇(TC)含量,Western blot检测脂代谢及炎症相关信号通路关键蛋白(PPARγ、NF-κB p65)的表达水平。[结果]:与空白对照组相比,PA模型组细胞内脂质滴显著增多,TG、TC含量显著升高(P<0.05),细胞上清液中IL-1β、IL-6分泌量显著增加(P<0.05),PPARγ表达水平下调而p-NF-κB p65蛋白表达水平显著上调(P<0.05);与PA模型组相比,LNT低剂量或高剂量干预组细胞内TG、TC含量显著降低,IL-1β、IL-6分泌量显著下降(P<0.05),PPARγ表达水平明显上调而p-NF-κB p65表达水平下调(P<0.05),且呈一定剂量依赖性。[结论]:LNT拮抗PA诱导的RAW264.7巨噬细胞脂质蓄积和炎症因子分泌,其作用机制可能与调控PPARγ/NF-κB信号通路相关。
Abstract
[Objective]: To investigate the antagonistic effects of lentinan (LNT) on lipid accumulation and inflammatory cytokine secretion in macrophages induced by palmitic acid (PA), and to provide experimental evidence for the application of LNT in metabolic inflammation-related diseases. [Methods]: RAW264.7 macrophages were divided into a control group, a PA model group, and low-/medium-/high-dose LNT intervention groups. Lipid accumulation and inflammatory models were established by treating RAW264.7 cells with different concentrations of PA, and the optimal modeling concentration was screened. In subsequent experiments, cells in the LNT groups were pretreated with corresponding concentrations of LNT, followed by co-incubation with the optimal concentration of PA. The secretion levels of interleukin-1 (IL-1) and interleukin-6 (IL-6) in the cell supernatant were determined by enzyme-linked immunosorbent assay (ELISA). Intracellular contents of triglyceride (TG) and total cholesterol (TC) were measured using biochemical kits. The expression levels of key proteins (PPAR, NF-B p65) in lipid metabolism and inflammation-related signaling pathways were detected by Western blot. [Results]: Compared with the control group, the PA model group showed significantly increased intracellular lipid droplets, elevated TG and TC contents (P<0.05), enhanced secretion of IL-1 and IL-6 in the supernatant (P<0.05), downregulated expression of PPAR, and upregulated expression of p-NF-B p65 (P<0.05). Compared with the PA model group, low- or high-dose LNT intervention significantly reduced intracellular TG and TC contents, decreased the secretion of IL-1 and IL-6 (P<0.05), upregulated PPAR expression, and downregulated p-NF-B p65 expression (P<0.05), in a dose-dependent manner. [Conclusion]: LNT antagonizes PA-induced lipid accumulation and inflammatory cytokine secretion in RAW264.7 macrophages. Its mechanism may be related to the regulation of the PPAR/NF-B signaling pathway.关键词
