Synthesis and application of a Novel Hydrogen Sulfide Fluorescent Probe Based on Nitroso
摘要
硫化氢(H2S)作为重要气体信号分子,在生物体内参与众多生理过程,其水平异常与多种疾病相关,其在食品安全领域也意义重大。亚硝基近红外区特征性强、基团还原性好,发射峰强度高、托克斯位移大的吡喃腈类骨架结合,设计合成了亚硝基苯基修饰的吡喃腈骨架的近红外荧光探针分子O-DCM-NO并进行结构表征确证。该探针利用亚硝基作为识别基团,基于硫化氢还原亚硝基芳基化合物为芳胺的反应机制,实现荧光"OFF-ON"识别硫化氢。通过对探针进行紫外光谱检测、时间响应研究、含水量研究、pH依赖性研究、荧光滴定研究、选择性研究以及RGB定量检测研究等进行分析,结果表明,该探针对H2S响应迅速,具有良好的选择性;在一定的HS-浓度范围呈线性关系,且肉眼可NaHS浓度和荧光颜色呈正相关。该探针有望为食品检测领域的H2S检测提供新方法,推动荧光探针技术在生物分析领域的发展。?????
Abstract
Hydrogen sulfide (H2S), as a crucial gaseous signaling molecule, participates in numerous physiological processes within organisms. Its abnormal levels are associated with various diseases and hold significant implications in food safety.The nitroso group exhibits strong characteristics in the near-infrared region, featuring excellent group reducibility, high emission peak intensity, and substantial Stokes shift when combined with a pyranonitrile skeleton. Based on these properties, a near-infrared fluorescent probe molecule, O-DCM-NO, featuring a nitrosophenyl-modified pyranonitrile skeleton, was designed, synthesized, and structurally characterized for confirmation.This probe utilizes the nitroso group as the recognition moiety, leveraging the reaction mechanism of H2S reducing nitrosoaryl compounds to arylamines, to achieve fluorescence "OFF-ON" recognition of hydrogen sulfide.Through analyses such as UV spectral detection, time-response studies, water content research, pH-dependence studies, fluorescence titration, selectivity studies, and RGB quantitative detection, the results demonstrate that this probe responds rapidly to H2S with excellent selectivity.Within a certain HS- concentration range, it exhibits a linear relationship, and the fluorescence color shows a positive correlation with NaHS concentration visible to the naked eye.This probe holds promise for providing a novel method for H2S detection in the food industry and advancing the development of fluorescent probe technology in bioanalytical applications.关键词
