Titanium Dioxide Nanotube-Based Local Delivery System Loaded with Cinobufagin

摘要
针对口腔癌治疗中易转移易复发的问题,构建了一种二氧化钛纳米管负载华蟾酥毒基的局部药物递送体系。华蟾酥毒基为中药蟾酥的主要活性提取物之一,具有抑制口腔癌细胞增殖的作用。采用阳极氧化法制备二氧化钛纳米管阵列,通过物理吸附法负载华蟾酥毒基。利用扫描电子显微镜表征纳米管形貌,通过傅里叶变换红外光谱与亲水角测试验证材料的成功制备及亲水性能,采用紫外分光光度法考察药物的负载与释放行为。结果表明,所制备的二氧化钛纳米管结构规整、排列有序;华蟾酥毒基成功负载于纳米管内并实现有效释放。该体系为华蟾酥毒基的局部递送及抑制口腔癌复发提供了可行的材料构建策略。
Abstract
To address the challenges of high recurrence and metastasis rates in oral cancer treatment, a local drug delivery system was developed using titanium dioxide nanotubes loaded with cinobufagin. Cinobufagin, one of the main active components extracted from the traditional Chinese medicine Bufonis Venenum exhibits inhibitory effects on the proliferation of oral cancer cells. Titanium dioxide nanotube arrays were fabricated via anodization, and cinobufagin was loaded through physical adsorption. The morphology of the nanotubes was characterized using scanning electron microscopy, while successful fabrication and hydrophilic properties were confirmed via Fourier-transform infrared spectroscopy and water contact angle measurements. Drug loading and release behaviors were evaluated using ultraviolet spectrophotometry. The results demonstrated that the prepared titanium dioxide nanotubes exhibited a well-ordered and uniform structure. Cinobufagin was successfully loaded into the nanotubes and achieved effective release. This system provides a feasible material-based strategy for the localized delivery of cinobufagin and the inhibition of oral cancer recurrence.关键词
