摘要

喹啉作为典型的含氮双环芳香杂环化合物,是许多生物活性天然产物和药物分子的关键组成部分,广泛存在于抗肿瘤、抗病毒等药物中。联烯醌(Vinylideneo-quinone methide)中间体作为高反应活性,多反应位点的手性试剂,被广泛应用于各种不对称反应中。本研究工作中,我们将联烯醌与5-取代喹啉环结合,合成一种结构新颖的含喹啉骨架的联烯醌前体化合物,丰富了联烯醌化学的应用范围,为后续构建喹啉新型和联烯醌的新型骨架化合物提供研究基础。

Abstract

Quinoline, a typical nitrogen-containing bicyclic aromatic heterocycle, serves as a key structural component in numerous bioactive natural products and pharmaceutical molecules, and is widely found in antitumor, antiviral and other therapeutic agents. As highly reactive and multisite chiral reagents, vinylidene ortho-quinone methide (VQM) intermediates have been widely employed in various asymmetric reactions. In this work, we integrated vinylidene ortho-quinone methides with 5-substituted quinoline rings to synthesize a series of novel quinoline-fused vinylidene ortho-quinone methide precursors. This study expands the application scope of vinylidene ortho-quinone methide chemistry and provides a foundation for the subsequent construction of novel skeletons involving quinoline and vinylidene ortho-quinone methide moieties.
