摘要

蛋白质药物因高特异性、低毒性的优势在肿瘤、遗传病等疾病治疗中展现出巨大潜力,但分子量大、膜穿透性差、易被酶降解等固有缺陷严重制约其临床转化,开发高效胞内蛋白递送载体成为关键突破口。本文系统综述了胞内蛋白递送的主要载体策略,包括病毒载体、脂质体递送系统、无机纳米材料、天然蛋白质载体及聚合物载体的结构特性、递送机制、应用现状与局限性,重点分析各类载体在蛋白负载、细胞摄取、内体逃逸等关键环节的性能表现,探讨不同载体策略的优化方向与发展趋势,为新型高效胞内蛋白递送系统的设计与开发提供参考。

Abstract

Protein drugs have shown great potential in the treatment of tumors, genetic diseases and other diseases due to their high specificity and low toxicity. However, inherent defects such as large molecular weight, poor membrane permeability and easy enzymatic degradation seriously restrict their clinical translation, making the development of efficient intracellular protein delivery carriers a key breakthrough. This paper systematically reviews the main carrier strategies for intracellular protein delivery, including the structural characteristics, delivery mechanisms, application status and limitations of viral vectors, liposome delivery systems, inorganic nanomaterials, natural protein carriers and polymer carriers. It focuses on analyzing the performance of various carriers in key links such as protein loading, cell uptake and endosomal escape, and discusses the optimization directions and development trends of different carrier strategies, providing a reference for the design and development of novel and efficient intracellular protein delivery systems.
