摘要

芳基C-糖苷(ACGs)因碳碳键结构稳定、口服生物利用度高,在抗生素、抗肿瘤及抗糖尿病药物研发中具有重要应用价值,备受合成与药物化学领域关注。糖基氯作为关键糖基供体,是构建糖苷骨架及制备烯糖等中间体的核心原料。本课题组长期致力于铁催化还原偶联构建芳基C-糖苷的研究,为拓展该催化体系的底物范围,本文建立了一种通用的糖基氯衍生物合成方法。该方法适用于2,3-位丙叉保护的五元环糖骨架,可高效制备苄基醚、硅醚、烷基醚及烷氧烷基醚等多种保护基修饰的糖基氯衍生物,为其后续功能化转化及芳基C-糖苷的合成研究提供了丰富底物基础。

Abstract

Aryl C-glycosides (ACGs) represent an important class of bioactive molecules with remarkable stability and excellent oral bioavailability, showing great promise in antibiotics, antitumor, and antidiabetic drug discovery. Glycosyl chlorides serve as versatile glycosyl donors and pivotal intermediates for the construction of glycoside frameworks.In ourcontinuous efforts toward iron-catalyzed reductive coupling for aryl C-glycosides, we herein describe a general synthetic method toward glycosyl chloride derivatives. This protocol is amenable to 2,3-O-isopropylidene-protected five-membered sugars and allows efficient preparation of diverse glycosyl chlorides bearing benzyl, silyl, alkyl, and alkoxyalkyl ether protecting groups. This work provides a valuable substrate platform for further functionalization and the synthesis of structurally diverse aryl C-glycosides.
