摘要

独脚金内酯(Strigolactones,SLs)是近年来被确认的一类重要植物激素,在植物生长发育及与环境互作中发挥关键调控作用,包括抑制植株分枝、促进植物与丛枝菌根真菌共生,以及诱导根际杂草种子萌发等,其功能实现依赖于其在植物体内及向根际环境的精准转运。ATP结合盒(ABC)G亚家族转运蛋白已在多种物种中被证实是介导包含SLs等多种植物激素跨膜转运的关键蛋白,但水稻中SLs转运蛋白至今仍未被系统鉴定。本研究旨在探究水稻一个与矮牵牛中SLs转运蛋白PaPDR1同源性较高的ABC成员-OsSLT1(strigolactone transporter)是否具有参与SLs转运的潜在生物学功能。为此本研究开展了以下研究采用亚细胞定位、酵母异源表达、三种实验证明了OsSLT1具有在水稻中转运植物激素SLs的潜在功能,它能够通过介导 SLs的根系分泌与体内运输,协同调控水稻的分蘖发育与抗寄生反应。该研究结果不仅丰富了植物激素转运的理论体系,也为通过分子育种手段同步改良水稻株型和培育抗寄生水稻品种提供了重要的候选基因与理论依据。

Abstract

Strigolactones (SLs) are a class of important plant hormones identified in recent years, playing key regulatory roles in plant growth, development, and plant-environment interactions. Their functions include inhibiting shoot branching, promoting symbiosis between plants and arbuscular mycorrhizal fungi, and inducing the germination of rhizosphere weed seeds. The realization of these functions depends on the precise transport of SLs within the plant and their secretion into the rhizosphere. ATP-binding cassette (ABC) G subfamily transporters have been confirmed in multiple species as key proteins mediating the transmembrane transport of various plant hormones, including SLs. However, SLs transporters in rice have not yet been systematically identified. This study aims to investigate whether OsSLT1 (strigolactone transporter), a rice ABC family member with high homology to the SLs transporter PaPDR1 from petunia, possesses potential biological functions involved in SLs transport. To this end, this study employed subcellular localization analysis, yeast heterologous expression, and other experimental approaches to demonstrate that OsSLT1 has the potential function of transporting the plant hormone SLs in rice, and that it may coordinately regulate rice tillering development and parasitic resistance by mediating the root secretion and internal transport of SLs. These findings not only enrich the theoretical framework of plant hormone transport but also provide important candidate genes and a theoretical basis for simultaneously improving rice plant architecture and breeding parasitic-resistant rice varieties through molecular breeding.
