摘要

胰腺癌预后极差,现有手术、化疗、靶向及免疫治疗疗效有限,亟需新型干预策略。天然产物可通过诱导凋亡、阻滞细胞周期、抑制促癌信号、调控肿瘤微环境及干预代谢重编程等多途径发挥抗肿瘤作用,显示出重要研发潜力。内质网应激(ERS)及未折叠蛋白反应(UPR)作为调控胰腺导管腺癌(PDAC)细胞命运的核心分子网络,是天然产物干预的关键靶点。本文系统综述ERS/UPR通路的分子机制及相关天然产物研究进展,并就生物利用度、作用机制与临床转化等核心问题进行探讨,以期为抗胰腺癌药物研发提供理论参考。

Abstract

Pancreatic cancer has an extremely poor prognosis, and the efficacy of existing treatments including surgery, chemotherapy, targeted therapy and immunotherapy is limited, highlighting an urgent need for novel intervention strategies. Natural products exert anti-tumor effects through multiple pathways, such as inducing apoptosis, arresting the cell cycle, inhibiting pro-tumorigenic signaling, regulating the tumor microenvironment, and interfering with metabolic reprogramming, thus demonstrating significant potential for drug development. Endoplasmic reticulum stress (ERS) and the unfolded protein response (UPR), as core molecular networks governing the cell fate of pancreatic ductal adenocarcinoma (PDAC), represent crucial targets for intervention by natural products. This paper systematically reviews the molecular mechanisms of the ERS/UPR pathway and advances in related natural product research, and discusses key issues including bioavailability, mechanism of action, and clinical translation, aiming to provide theoretical references for the development of anti-pancreatic cancer drugs.
