摘要

随着化学杀线剂的逐步禁用,开发和利用环境友好、安全高效的生防菌已成为植物病害防控领域的迫切需求。本文利用96孔板筛选获得一株具有显著杀线活性的根际细菌--烟嘧磺隆假单胞菌(Pseudomonas nicosulfuronedens)MiC45,并对其活性物质进行了分析和研究。盆栽实验显示,菌株MiC45发酵液处理能显著降低辣椒根系中线虫数、根结数,延缓线虫发育,并能显著促进辣椒的生长。利用液相色谱-质谱联用技术(LC-MS),从MiC45发酵液的次级代谢产物中分离、鉴定出四种具有杀线虫活性的有机酸:反式乌头酸(trans-aconitic acid)、壬二酸(azelaic acid)、辛二酸(suberic acid)和对香豆酸(p-coumaric acid)。其中,壬二酸对线虫致死率最高,处理24 h后达88.36%,并诱导线虫体内形成空泡样结构。本文揭示了P. nicosulfuronedens MiC45及其活性成分壬二酸在生物防治方面的巨大潜力。

Abstract

The progressive phase-out of chemical nematicides has necessitated the urgent development of environmentally benign, safe, and efficient biocontrol agents for plant disease management. In this study, the potent nematicidal rhizosphere bacterium Pseudomonas nicosulfuronedens MiC45 was isolated using a 96-well plate, and its bioactive metabolites were characterized. Pot experiments demonstrated that treatment with the MiC45 fermentation broth significantly reduced nematode populations and root galling in pepper plants, delayed nematode development, and markedly promoted plant growth. Liquid chromatography-mass spectrometry (LC-MS) analysis identified four organic acids with nematicidal activity from the secondary metabolites of the MiC45 fermentation brIdentification and nematicidal activity of biocontrol strain MiC45 against Meloidogyne incognitaoth: trans-aconitic acid, azelaic acid, suberic acid, and p-coumaric acid. Among these, azelaic acid exhibited the most potent nematicidal activity, achieving a mortality rate of 88.36% after 24 hours of treatment and inducing the formation of vacuole-like structures within the nematodes. These findings highlight the significant potential of P. nicosulfuronedens MiC45 and its bioactive component, azelaic acid, as promising candidates for biological control.
