CDK7调控结直肠癌进展的初步研究

摘要
结直肠癌（colorectal cancer，CRC）是消化系统常见的恶性肿瘤之一，具有较高的发病率和死亡率，其发生发展与多种基因异常表达及恶性生物学行为改变密切相关。为探讨细胞周期蛋白依赖性激酶7（cyclin-dependent kinase 7，CDK7）在结直肠癌中的作用，本研究以结直肠癌细胞为研究对象，初步分析CDK7对肿瘤细胞增殖与迁移能力的影响。首先，通过生物信息学方法分析CDK7在结直肠癌组织及正常组织中的表达差异，结果显示CDK7在结直肠癌组织中呈高表达。进一步结合蛋白免疫印迹实验，对CDK7的蛋白表达水平进行了验证。随后，通过构建CDK7过表达细胞模型，采用EdU实验评价细胞DNA复制水平，利用划痕愈合实验检测细胞迁移能力。研究结果表明，CDK7过表达可显著增强结直肠癌细胞的增殖能力和迁移能力，表现为EdU阳性细胞比例增加以及划痕愈合速度加快。上述结果表明，CDK7在结直肠癌细胞恶性生物学行为中发挥促进作用，可能参与结直肠癌的发生与发展，并有望成为结直肠癌诊断与治疗的潜在分子靶点。
Abstract
Colorectal cancer (CRC) is one of the most common malignant tumors of the digestive system, with high incidence and mortality rates. Its occurrence and progression are closely associated with abnormal expression of multiple genes and alterations in malignant biological behaviors. To investigate the role of cyclin-dependent kinase 7 (CDK7) in colorectal cancer, this study used colorectal cancer cells as the research model to preliminarily analyze the effects of CDK7 on tumor cell proliferation and migration. First, bioinformatics analysis was performed to evaluate the differential expression of CDK7 between colorectal cancer tissues and normal tissues, and the results showed that CDK7 was highly expressed in colorectal cancer tissues. Western blot analysis was then conducted to further verify the protein expression level of CDK7. Subsequently, a CDK7-overexpressing cell model was established, and EdU assays were performed to assess DNA replication activity, while wound healing assays were used to evaluate cell migratory ability. The results demonstrated that CDK7 overexpression significantly enhanced the proliferative and migratory capacities of colorectal cancer cells, as evidenced by an increased proportion of EdU-positive cells and accelerated wound closure. These findings indicate that CDK7 plays a promoting role in the malignant biological behavior of colorectal cancer cells, may be involved in the occurrence and progression of colorectal cancer, and could serve as a potential molecular target for the diagnosis and treatment of colorectal cancer.关键词
