摘要
本研究围绕本研究旨在构建并优化光滑念珠菌铜离子诱导型表达体系,以实现外源蛋白的高效可控表达。方法上,首先对金属硫蛋白启动子MT1和MT2在不同培养基(SD-Ura与YPD)及不同CuSO?浓度条件下的诱导特性进行系统分析,确定其适宜诱导范围;随后构建表达载体,以mEGFP为报告蛋白评估体系的表达能力与可控性。结果表明,MT1启动子整体诱导性能优于MT2,且对铜离子呈良好的剂量依赖性响应。并纯化了mEGFP,验证该体系在蛋白表达研究中的适用性。结果证明在该启动子调控下,可实现铜离子依赖的可控表达,获得功能正常的蛋白。本研究建立了一种在光滑念珠菌中具有良好可控性和较高表达效率的铜离子诱导型表达体系。
Abstract
This study aimed to construct and optimize a copper ion-inducible expression system in Candida glabrata for efficient and controllable heterologous protein expression. First, the inducible properties of the metallothionein promoters MT1 and MT2 were systematically analyzed under different culture conditions (SD-Ura and YPD media) and varying CuSO? concentrations to determine their optimal induction ranges. Subsequently, expression vectors were constructed, and mEGFP was used as a reporter protein to evaluate the expression capacity and regulatory performance of the system. The results showed that the MT1 promoter exhibited overall superior inducible performance compared with MT2 and displayed a strong dose-dependent response to copper ions.Furthermore, mEGFP was successfully purified to validate the applicability of this system for protein expression studies. The results demonstrated that copper-dependent controllable expression could be achieved under promoter regulation, yielding functionally intact protein. In conclusion, this study established a copper ion-inducible expression system in C. glabrata with good controllability and relatively high expression efficiency.关键词
