摘要

糖苷水解酶(GHs,Glycoside hydrolases)是水解糖苷键的酶,在生物体中糖和糖苷化合物的降解和合成中发挥着关键作用。根据碳水化合物活性酶数据库(CAZy,Carbohydrate active enzyme)分类,GHs分为189个家族(糖苷水解酶家族),编号从GH1到GH189,每个家族的成员具有高度的结构相似性,GH17家族酶在植物中具有多种功能。【目的】为了探究GH17基因家族在棉花中的存在形式和功能,本研究利用生物信息学从陆地棉全基因组中鉴定出132个GH17基因,并对其蛋白质理化特性、基因结构、系统发育树、顺势作用元件、表达模式和Ka/Ks比值进行分析。【方法】通过不同软件鉴定GH17基因家族成员,并对其进行生物信息学分析。【结果】结果表明,132个陆地棉GH17基因家族成员分为6个亚族(Ⅰ~Ⅵ),较多分布在A/D05、A/D06、A/D07、A/D08、A/D09、A/D10条等染色体上。棉花GH17基因家族的编码区长度为312~2628 bp;氨基酸长度为103~875 aa;蛋白分子量为11 656.53~95 438.15 kD;蛋白等电点为4.23~9.89。陆地棉GH17基因家族的亚细胞定位预测结果显示,该家族主要定位在叶绿体、细胞膜、液泡内,部分定位在内质网、细胞核、细胞质和细胞外中。此外,GH17基因家族成员在不同组织,不同时期表现出不同的表达模式,其在根、茎和雄蕊器官中表达量较高;部分GhGH17基因能够响应冷处理、热处理、干旱胁迫、盐胁迫等多种非生物胁迫。【结论】本研究为陆地棉GH17基因家族的生物学功能和棉花抗逆基因的研究提供了一定的参考。

Abstract

Glycoside hydrolases (GHs), enzymes that hydrolyze glycosidic bonds, play a crucial role in the degradation and synthesis of sugars and glycosides within organisms. According to the Carbohydrate Active Enzymes (CAZy) database classification, GHs are categorized into 189 families (Glycoside Hydrolase Families), numbered from GH1 to GH189, with members within each family exhibiting high structural similarity. GH17 family enzymes perform diverse functions in plants.【Objective】To investigate the presence and potential functions of GH17 genes in cotton, this study identified 132 GH17 genes from the whole genome of upland cotton using bioinformatics methods, followed by analyses of their protein physicochemical properties, gene structure, phylogenetic trees, cis-acting elements, expression patterns, and Ka/Ks specific value.【Methods】The GH17 gene family members were identified by different software and bioinformatics analysis was performed.【Result】Results showed that the 132 land cotton GH17 genes family members are divided into six subfamilies (I-VI), predominantly distributed across chromosomes A/D05, A/D06, A/D07, A/D08, A/D09, and A/D10. The coding regions of the cotton GH17 gene family range from 312 to 2628 bp, with amino acid lengths spanning 103 to 875 aa. Their protein molecular weights range from 11,656.53 to 95,438.15 kDa, and the protein isoelectric points are between 4.23 and 9.89. Subcellular localization predictions indicate that this gene family is primarily localized in chloroplasts, cell membranes, and vacuoles, with partial distribution in the endoplasmic reticulum, nucleus, cytoplasm, and extracellular matrix. Additionally, GH17 gene family members expression patterns vary across tissues and developmental stages, showing higher abundance in roots, stems, and stamens. Some GhGH17 genes exhibit stress-responsive mechanisms to cold treatment, heat stress, drought, and salt stress.【Conclusion】This study provides valuable insights for understanding the biological functions of the land cotton GH17 gene family and advancing research on stress-resistant genes in cotton.
