摘要

目的:本研究旨在通过优化γδT细胞制备和扩增流程,构建基于CD33纳米抗体的CAR-γδT细胞,通过体外实验评估其抗肿瘤活性。方法:通过慢病毒转染构建四种CD33?CAR?γδT细胞,使用流式细胞术检测转染效率、体外抗肿瘤活性、细胞因子释放、分化表型以及扩增能力进行检测。结果:制备高滴度的慢病毒,成功构建CD33-CAR-γδT细胞,在体外验证其具有良好的抗肿瘤活性。结论:本研究成功构建四种结构的CD33-CAR-γδT细胞,四种细胞均具备体外抗肿瘤活性,其中BC结构表现最优,在记忆表型维持(高TNaive比例)及持续杀伤能力上显著优于其他结构。

Abstract

Objective: This study aimed to construct CD33 nanobody-based CAR?γδT cells by optimizing the preparation and expansion protocols of γδT cells, and to evaluate their anti?tumor activity in vitro. Methods: Four types of CD33-CAR-γδT cells were constructed via lentiviral transfection, and flow cytometry was performed to detect transfection efficiency, in vitro anti?tumor activity, cytokine release, differentiation phenotype, and expansion capacity. Results: High-titer lentiviruses were prepared, and CD33-CAR-γδT cells were successfully constructed and verified to exhibit favorable anti-tumor activity in vitro. Conclusion: Four structurally distinct CD33-CAR-γδT cells were successfully established in this study, all of which exerted in vitro anti?tumor activity; among them, construct BC showed the optimal performance, with memory phenotype maintenance (high proportion of naive T cells), and sustained cytotoxicity compared with the other structures.
