摘要

胞间液在植物信号转导过程中发挥着多种重要作用。目前尚无关于小麦叶片胞间液提取方法的报道。本研究建立并优化了提取小麦胞间液的真空渗透离心法。以苏麦、扬麦和Fielder小麦品种为试验材料,该改良VIC法仅利用普通实验室易获取的器材,如注射器、保鲜膜、凡士林、移液枪头和离心管即可完成。通过对蛋白含量、叶绿体污染程度、细胞损伤水平及叶片相对电导率进行综合评价,确定在500 rcf离心条件下,1 g小麦叶片组织适用于上述3个小麦品种幼苗的胞间液提取。

Abstract

IApoplast fluid (APF) is playing multiple important roles in signaling transduction in plants. However, there was no report showing how to extract APF from wheat leaf. In this study, a modify and optimize the vacuum infiltration/centrifugation (VIC) for APF extraction in wheat was developed and carefully described below. Sumai, Yangmai and Fielder wheat cultivars were used as plant material. Modified VIC method for wheat is only using easy access material in normal laboratory, such as syringe, cling film, Vaseline, pipette tip and centrifuge tube. To optimize the VIC method, the protein level, chloroplast contamination, cell damage level and relative conductivity of leaf segment was evaluated. And according to the data, 1g leaf tissue under 500 rcf centrifugation level is suitable for APF extraction in all three wheat variety seedlings.
