陆地棉CSE基因家族鉴定及表达分析

摘要
咖啡酰莽草酸酯酶(Caffeoylshikimate esterase,CSE)是植物木质素合成途径中的关键限速酶,在棉花细胞壁生长及细胞伸长活动中发挥重要作用。为解析棉花CSE基因家族的进化特征及其在陆地棉中的潜在功能,本研究利用生物信息学技术,鉴定了陆地棉、海岛棉、亚洲棉和雷蒙德氏棉4个棉种中的CSE基因家族成员并构建系统进化树,同时分析了陆地棉中CSE基因家族成员的理化性质、启动子顺式作用元件及其发育早期组织表达模式。结果表明,在4个棉种内共含有112个CSE家族成员,包括38个陆地棉GhCSE基因,37个海岛棉GbCSE基因,19个亚洲棉GaCSE基因和18个雷蒙德氏棉GrCSE基因,系统进化将其分为6个亚组。陆地棉CSE家族成员的蛋白整体偏碱性,大部分为稳定蛋白,亚细胞定位预测结果主要为细胞质。顺式作用元件分析预测显示大部分元件与光响应和逆境胁迫相关。基于出苗时期转录组数据,对陆地棉38个CSE基因的组织表达量进行分析,发现大部分CSE基因在出苗表现优异材料中表达量显著高于出苗表现不佳材料,且主要在下胚轴中优势表达,初步筛选出GhCSE14、GhCSE32等调控棉花出苗的候选基因,为深入研究CSE基因在棉花早期发育中的功能奠定理论基础。
Abstract
Caffeoylshikimate esterase (CSE) is a key rate-limiting enzyme in the plant lignin biosynthesis pathway, and it plays an essential role in cotton cell wall growth and cell elongation. To dissect the evolutionary characteristics of the cotton CSE gene family and its potential functions in upland cotton, bioinformatics methods were used in this study to identify the members of the CSE gene family in four cotton species, including upland cotton, island cotton, Asiatic cotton, and Raymond\'s cotton. A phylogenetic tree was constructed, and the physicochemical properties, promoter cis?acting elements, and tissue expression patterns during early seedling development of the CSE gene family members in upland cotton were systematically analyzed. The results showed that a total of 112 CSE family members were identified in the four cotton species, including 38 GhCSE genes in upland cotton, 37 GbCSE genes in island cotton, 19 GaCSE genes in Asiatic cotton, and 18 GrCSE genes in Raymond\'s cotton. All CSE genes were classified into six subgroups according to phylogenetic analysis. Protein property analysis indicated that most GhCSE proteins were weakly alkaline and stable, and subcellular localization prediction showed that they were mainly located in the cytoplasm. Cis?acting element analysis revealed that most promoter elements were related to light response and stress response. Based on the transcriptome data during the seedling emergence stage, the expression patterns of 38 GhCSE genes in upland cotton were further analyzed. The results showed that the expression levels of most GhCSE genes were significantly higher in materials with excellent seedling emergence performance than in those with poor seedling emergence performance, and were predominantly expressed in hypocotyl tissues. Among them, GhCSE14 and GhCSE32 were preliminarily screened as candidate genes regulating cotton seedling emergence. This study provides a theoretical foundation for further functional research of CSE genes in cotton early development.关键词
