摘要

为了探究AgRP神经元中Gpr45基因对高脂饮食诱导的雄性小鼠能量代谢的调控作用,利用Cre-loxP系统构建AgRP神经元特异性Gpr45基因敲除小鼠,给予8周60%的高脂饮食喂养,监测体重、体成分、摄食量、能量消耗及糖代谢状态,检测血浆生化指标及肝脏组织学改变。研究结果表明AgRP神经元中Gpr45基因缺失可加剧高脂饮食诱导的雄性小鼠肥胖、胰岛素抵抗及肝脂肪变性,说明该基因在维持能量平衡及肝脏代谢稳态中发挥一定作用。

Abstract

To investigate the role of Gpr45 in AgRP neurons in regulating energy metabolism in male mice under high-fat diet conditions, an AgRP neuron-specific Gpr45 knockout mouse model was generated using the Cre-loxP system. Mice were fed a 60% high-fat diet for 8 weeks, during which body weight, body composition, food intake, energy expenditure, and glucose metabolism were systematically assessed. In addition, plasma biochemical parameters and liver histopathology were analyzed. The results demonstrated that deletion of Gpr45 in AgRP neurons exacerbated high-fat diet-induced obesity, insulin resistance, and hepatic steatosis in male mice, suggesting that Gpr45 in AgRP neurons plays a critical role in maintaining energy homeostasis and hepatic metabolic balance.
