摘要

植物KASⅠ基因在脂肪酸合成过程中具有重要作用,为鉴定棉花KASⅠ基因家族并明确KASⅠ的表达特征,利用生物信息学手段对二倍体和四倍体棉种中KASⅠ基因家族进行分析,并进一步结合转录组数据和RT-qPCR分析了KASⅠ基因在纤维发育时期的表达特征。结果表明,在陆地棉与海岛棉中共鉴定出12个KASⅠ基因家族成员,根据系统发育进化关系分为3个亚族,部分基因在纤维伸长期特异表达。荧光定量和转录组测序结果表明,GhKASⅠ_A09与GbKASⅠ_A09在陆地棉与海岛棉中差异表达,对GhKASⅠ_A09与GbKASⅠ_A09的CDS序列分析表明,陆地棉与海岛棉的KASⅠ序列之间均存在4 bp的Indel差异。上述结果为进一步分析棉花KASⅠ家族基因以及分子机制提供了一定的理论基础。

Abstract

The plant KASⅠ gene plays an essential role in fatty acid biosynthesis. To identify the KASⅠ gene family in cotton and clarify its expression patterns, bioinformatics approaches were used to analyze the KASⅠ gene family in diploid and tetraploid cotton species. Furthermore, transcriptome data and RT-qPCR were employed to characterize the expression profiles of KASⅠ genes during cotton fiber development. The results showed that a total of 12 KASⅠ family members were identified in upland cotton and island cotton, which were classified into three subfamilies based on phylogenetic relationships, and some genes exhibited specific expression during the fiber elongation stage. RT-qPCR and transcriptome sequencing results revealed differential expression of GhKASⅠ_A09 and GbKASⅠ_A09 between upland cotton and island cotton. CDS sequence analysis of GhKASⅠ_A09 and GbKASⅠ_A09 identified a 4 bp Indel variation in the KASⅠ sequences of the two cotton species. These findings provide a theoretical foundation for further exploration of the functions and molecular mechanisms of the cotton KASⅠ gene family.
