摘要

马兜铃酸(Aristolochic acids,简称AAs)是广泛存在于马兜铃科马兜铃属Aristolochia 植物的一类硝基菲有机酸类化合物,具有强肾毒性、致突变性和致癌性,被世界卫生组织列为 Ⅰ 类致癌物,其中马兜铃酸 Ⅰ(AA Ⅰ)是毒性最强的核心组分。因此,为开发高效的 AA Ⅰ 检测探针,本研究基于噬菌体展示技术平台,利用全合成亲合体分子库,通过多轮富集淘选出马兜铃酸Ⅰ特异性结合亲合体。利用BL21(DE3)宿主细胞表达系统制备马兜铃酸Ⅰ特异性结合亲合体,并结合荧光淬灭实验验证淘选到的特异结合亲合体与马兜铃酸Ⅰ的结合能力及结合特异性。结果表明,筛选获得的亲合体 A5 可与 AA Ⅰ 发生特异性结合,二者结合过程主要依赖氢键和范德华力,且该亲合体具有制备成本低、操作简便的优势。本研究为中药材、天然植物及食品中 AA Ⅰ 的快速检测提供了新型识别探针,也为后续 AA Ⅰ 靶向吸附材料的开发奠定实验基础。

Abstract

Aristolochic acids (AAs) are a class of nitrophenanthrene organic acid compounds widely found in plants of the genus Aristolochia in the family Aristolochiaceae. They possess strong nephrotoxicity, mutagenicity, and carcinogenicity and are classified as Group I carcinogens by the World Health Organization, among which Aristolochic Acid I (AA I) is the most toxic core component. Therefore, to develop an efficient AA I detection probe, this study utilized a phage display technology platform and a fully synthetic affibody peptide library to identify peptides specifically binding to Aristolochic Acid I through multiple rounds of enrichment and selection. The AA I-specific binding peptides were prepared using a BL21(DE3) host cell expression system, and fluorescence quenching experiments were used to verify the binding ability and specificity of the selected peptides to AA I. The results showed that the selected affibody A5 can specifically bind to AA I, with the binding process mainly relying on hydrogen bonds and van der Waals forces. Furthermore, this affibody has the advantages of low preparation cost and ease of operation. This study provides a novel recognition probe for the rapid detection of AA I in Chinese herbal medicines, natural plants, and food, and lays an experimental foundation for the subsequent development of AA I-targeted adsorption materials.
