Theoretical design of esophageal cancer targeted nanomedicine based on effective components of Niuhuang Jiedu Tablets and its combined therapy: A Review
Esophageal cancer is a highly prevalent malignant digestive tract tumor in China. Traditional surgery, radiotherapy, and chemotherapy have certain limitations, making targeted therapy and immunotherapy research hotspots. Drawing inspiration from traditional monotherapy, our team proposes a novel targeted nanomedicine "LDXD" and its combined therapy theory design based on the effective components dimethyl arsenate and arsenic trioxide from Niuhuang Jiedu Tablets, combined with the "Jun Chen Zuo Shi" (emperor, minister, assistant, and messenger) compatibility theory of traditional Chinese medicine. This drug utilizes a liposomal nanocarrier surface-modified with esophageal squamous cell carcinoma-specific adhesion proteins to achieve precise targeting and membrane fusion drug delivery. In the combined therapy, the injection part includes LDXD (emperor drug), platycodon extract (minister drug), and berberine (assistant drug), while the oral part uses licorice and mung bean as "messenger drugs" to enhance drug targeting, reduce toxic side effects, and improve patient quality of life. The article also discusses the potential challenges this therapy may face in clinical translation and looks forward to the potential of artificial intelligence and nanotechnology in drug design, aiming to provide new ideas for the integrated traditional Chinese and Western medicine treatment of esophageal cancer.
