痘苗病毒是一种理想的基因治疗的载体,在肿瘤的基因治疗中有广泛的前景。重组痘苗病毒缺失了TK区,使其具有靶向性和溶瘤性,可选择性地感染肿瘤组织,使目的基因能较好地在肿瘤局部表达。本研究中将锰超氧化物歧化酶(MnSOD)基因插痘苗病毒TK基因区,构建成MnSOD"武装"化的重组溶瘤痘苗病毒(rVV-MnSOD),并研究其对HeLa肿瘤后研究对HeLa细胞的杀伤作用,采用不同浓度梯度和时间梯度,通过电子显微镜观察、 MTT检测、结晶紫染色检测细胞的存活率、Western blot检测重组痘苗病毒rVV-MnSOD感染HeLa细胞后的形态学变化、细胞存活率、细胞凋亡、相关蛋白质表达。结果显示,rVV-MnSOD能明显抑制HeLa细胞生长,目的蛋白MnSOD能够高效的表达,促使细胞的凋亡。

IVaccinia virus is a ideal gene therapy vector and has great potential in cancer gene therapy, Recombinant vaccinia virus has deleted TK, it can tumor-selective replication and lyse cancer cells, can selectively tumor tissue infection, purpose gene can express well in special area of tumor. This paper study killing-effect on HeLa by MnSOD gene-armed recombinant oncolytic vaccinia virus,which manganese superoxide dismutase ( MnSOD ) gene insert into TK gene of vaccinia virus. After treated with different-concentration of rVV-MnSOD at different-time, the change of morphological change, cells viability, cell apoptosis, the protein expression of related genes formation of in HeLa cells were assessed by fluorescence microscope, MTT assay, crystal violet assay , Western blotting and transmission electron microscopy analysis. The results suggested that rVV-MnSOD can inhibit HeLa cells growth remarkably. purpose gene can express well,consequently induced releasing of cytochrome c ,inducing apoptosis at last.
