Effect of high-molecular weight polycyclic aromatic hydrocarbons on the microbial communities in soils from vadose zone

地下水有机污染问题日益严重,我国很多地方的地下水中相继检出多环芳烃(PAHs)的存在,包括以荧蒽和芘为代表的4环及以上的高分子量PAHs。本研究采集长期受到原油污染的包气带土壤样品,以芘和荧蒽为碳源和能源在无机盐培养基中进行降解微生物的驯化,采用分子生物学方法,通过16S rDNA序列的分析来对比驯化前后微生物的群落特征。结果表明,该原油污染包气带土壤中的微生物具有较好的多样性,且可能存在一种未经鉴定的新菌种;以荧蒽和芘为碳源进行驯化后,土壤中的微生物种属由11种降至4种,分别为Pseudomonas 、Enterobacter、Hydrogenophaga和Luteolibacter pohnpeiensis。研究可为地下水的污染控制及生物修复研究提供一定的科学依据与参考。

Organic pollution in groundwater is getting worse. In China, polycyclic aromatic hydrocarbons (PAHs) are widespread in the groundwater, including high-molecular weight PAHs (HMW PAHs) like fluoranthene and pyrene. Soil samples, collected from vadose zone that has been polluted by crude oil for long time, was cultured in a mineral salt medium using fluoranthene and pyrene as the carbon and energy source for the selection of possible degrading bacteria. The microbial diversity and the relation of genus and species was analyzed adopting 16S rDNA sequencing method. Experimental results showed that the soil from vadose zone suffering from long-period crude oil pollution had a good microbial diversity. Besides, a bacterium belonging to an unknown genus was probably found. After incubation with fluoranthene and pyrene, the consortium in soils decreased to 4 species: Pseudomonas, Enterobacter, Hydrogenophaga and Luteolibacter pohnpeiensis. Results might provide scientifical basis for the further studies on the control and bioremediation of groundwater contamination.
