建立适用于体外培养的K562细胞总蛋白的双向电泳图谱,提高电泳分辨率和重复性,以进一步了解中药墓头回作用于白血病细胞的新机制。采用标准裂解法提取K562细胞总蛋白,后续进行丙酮沉淀和三氯乙酸 /丙酮沉淀,分别比较一步裂解法所得蛋白和丙酮沉淀法、三氯乙酸 /丙酮沉淀法所得蛋白的双向电泳图谱,优选最佳的样品制备方法。丙酮沉淀法不仅能减少蛋白降解,而且增加蛋白的溶解性,因此丙酮沉淀法所得蛋白的双向电泳图谱显示更好的溶解性和重复性,此外在双向凝胶电泳(2-DE)时,聚焦电压在传统聚焦条件上进行优化,成功建立了背景清晰,蛋白分离良好的白血病K562细胞的双向电泳体系。在聚焦条件上对传统聚焦条件的优化使横向和纵向拖尾有明显改善,所得图谱蛋白点更清晰,分离更完全,能获得分辨率较高的双向电泳图谱。在标准裂解液提取蛋白基础上结合丙酮沉淀法更适于制备K562细胞的双向电泳样品,等电聚焦(IEF)电泳时适当延长除盐时间能更有效的分离K562细胞全蛋白,而且后续实验证明此方法同样适用于肿瘤组织蛋白质的提取。

o optimize K562 cell sample preparation, optimal parameters of two-dimensional electrophoresis were established to improve resolution and reproducibility which is used to investigate the mechanism between diversifolious patrinia root and K562 cell. Standard lysis buffer were used to preparing K562 cell samples. Two different preparation methods, TCA/ Acetone precipitation and acetone precipitation, were applied to K562 cell samples and the acetone precipitation method was proved to be the better one considering the resheults obtained from two-dimensional gel electrophoresis (2-DE). Acetone precipitation not only decreased protein decomposition,but also increased protein solubility,therefore obtaining the two dimensional electrophoresis spectrum with higher resolution and better reproducibility. Besides, the focus voltage for gel electrophoresis was optimized on the traditional focus conditions and a two-dimensional electrophoresis system with a clear background and good protein separation was successfully established for leukemia K562 cells. The optimized focusing condition made horizontal and vertical tail improved obviously, and a high-resolution two-dimensional electrophoresis map with clearer protein spots and more complete separation obtained easily. Acetone precipitation on the basic of standard lysis buffer was more suitable for preparing the sample of K562 cell to operate two-dimensional electrophoresis. In addition, the longer desalinization time in isoelectric focusing (IEF) can separate whole protein effectively and the Following tests forcefully proves the wide utility of the optimized methods for the tumor tissues .
