香鱼巨噬细胞对鳗弧菌的吞噬作用

香鱼(Plecoglossus altivelis)是一种名贵的经济鱼类。本研究建立香鱼巨噬细胞培养,检测鳗弧菌对巨噬细胞呼吸爆发和噬菌能力影响。巨噬细胞由头肾组织经梯度离心,贴壁分离来获得,并通过姬姆萨染色证实。进一步的研究发现鳗弧菌(Listonella anguillarum)可以刺激香鱼巨噬细胞产生呼吸爆发,其还原NBT的能力比静息状态显著性增高。鳗弧菌加入到培养的巨噬细胞,在4 h香鱼巨噬细胞大量吞噬细菌,在12 h香鱼巨噬细胞内含的细菌减少。这是我国香鱼巨噬细胞体外培养的首次报道,香鱼巨噬细胞对鳗弧菌有很强杀伤能力,是抵抗病原体侵染的重要防线。
yu, Plecoglossus altivelis, is a popular and highly valued edible fish. We established primary culture of head kidney-derived macrophages of ayu. Moreover, the phagocytic capability and respiratory burst response of macrophages were observed when these cells were treated with Listonella anguillarum. The macrophages were obtained from head kidney of ayu by Ficoll gradient centrifugation, and removing non-adherent cells. The Giemsa staining was employed to confirm the morphology of ayu macrophages. The respiratory burst was estimated by measuring the intracellular O2- using the NBT reduction test. The results showed that the capability of ayu macrophages to generate a respiratory burst upon stimulation of Listonella anguillarum was higher than control. After adding Listonella anguillarum to cell media, an initial increase in the number of bacteria in the macrophages was seen at 4 h post-infection; a pronounced decrease was seen at 12 h post-infection. Our results suggest that ayu macrophages have the capability to kill Listonella anguillarum, which is an important host defense mechanism for Listonella anguillarum infected ayu.
